目的:对内蒙古阴山地区栽培和野生小秦艽的品质进行评价.方法:通过性状鉴别及薄层色谱法对小秦艽进行鉴别,采用液相色谱法对小秦艽主要成分的含量进行测定,采用液相色谱-质谱法对小秦艽中主要的化合物进行表征,结合主成分分析、热图等方法对结果进行分析.结果:小秦艽根部随生长年限增长逐渐出现分支和黑皮,颜色不断加深,直径和长度不断增加.薄层色谱结果显示,3~7 年生栽培小秦艽及野生小秦艽样品相应暗斑和显色与对照品一致,14 批小秦艽根中马钱苷酸质量分数为 0.233 2%~1.152 7%、龙胆苦苷质量分数为 4.688 9%~8.531 2%,总质量分数为 5.379 7%~9.043 8%,所有样地栽培小秦艽根中 2 个成分含量均达到《中华人民共和国药典》2020 年版标准.表征了不同生长年限小秦艽中以环烯醚萜为主的 20 个共有化学成分,并进行了相对定量.随着生长时间的延长,小秦艽中活性成分的积累逐年增长,但栽培 5 年后活性成分的积累趋缓,不同种植方法及不同种植密度对小秦艽的品质影响较小.结论:内蒙古阴山地区适合小秦艽的种植栽培,建议对生长期为 4~5 年的小秦艽进行采收及产地加工,所得药材质量较好.

Objective:To evaluate the quality of wild and cultivated Gentiana dahurica from Yinshan area of Inner Mongolia.Methods:Morphological characterization and thin-layer chromatography were employed to identify G.dahurica.High performance liquid chromatography(HPLC)was employed to determine the content of loganic acid and gentiopicroside.Liquid chromatography-mass spectrometry was employed to characterize the main chemical components in G.dahurica,and principal component analysis and heatmap analysis were performed to analyze the obtained data.Results:During the growing process,the roots of G.dahurica gradually showed branches and darken skin,with ncreases in diameter and length.Thin-layer chromatography showed that the dark spots and color of cultivated and wild G.dahurica of 3 to 7 years old were consistent with the reference standard.The mass fractions of gentiopicroside and loganic acid in 14 batches of G.dahurica samples were 4.688 9%-8.531 2%and 0.233 2%-1.152 7%,respectively.The total mass fractions of gentiopicroside and loganic acid in these samples were 5.379 7%-9.043 8%.The content of the two compomemts in the samples from all the plots met the requirements of the Chinese Pharmacopoeia(2020 version).The chemical composition of G.dahurica from different growth years was analyzed,which revealed a total of 20 common components,predominantly iridoids.Furthermore,relative quantification was conducted for these components.With the extension of growing time,the accumulation of active components in G.dahurica increased year by year,while the increase slowed down after 5 years of cultivation.Different planting methods and planting densities had little effect on the quality of G.dahurica.Conclusion:Yinshan area of Inner Mongolia is suitable for the cultivation of G.dahurica.It is suggested that G.dahurica of 4-5 years should be harvested and processed in the producing area,and the obtained medicinal materials demonstrate high quality.
