通过斑马鱼模型探究多酚类物质抗辣木叶蛋白致敏作用OA
Exploring the anti-allergic effects of polyphenols against the proteins from Moringa oleifera leaves using zebrafish model
为探究辣木叶蛋白的致敏作用以及多酚类物质如荷叶多酚和黄精多酚的抗过敏作用,本研究提取辣木叶蛋白(Moringa oleifera leaf protein,MLP)构建斑马鱼食物过敏模型,同时给予荷叶多酚提取物(lotus leaf polyphenol extract,LPE)和黄精多酚提取物(Polygonatum polyphenol extract,PPE)干预,通过行为学检测、肠道切片染色和实时荧光定量聚合酶链反应(real time fluorescent quantitative polymerase chain reaction,qRT-PCR)等方法,研究辣木叶蛋白的致敏作用及荷叶多酚和黄精多酚的抗过敏效果.结果表明,辣木叶蛋白降低了斑马鱼的运动能力,而荷叶多酚和黄精多酚能恢复这一指标,显示出保护作用.辣木叶蛋白对肠道屏障有显著损伤,而荷叶多酚和黄精多酚能加强免疫应答,促进肠道恢复,缓解过敏症状.qRT-PCR结果显示,荷叶多酚和黄精多酚通过调控炎症因子白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1β(IL-1β)、白细胞介素-4/13B(IL-4/13B)、白细胞介素-10(IL-10)等的表达,抑制辣木叶蛋白引起的炎症反应.本研究证实了辣木叶蛋白的致敏性及荷叶多酚和黄精多酚的抗过敏潜力,为食物过敏的预防和治疗提供了一定科学依据.
To explore the allergenic effects of Moringa oleifera leaf protein(MLP)and the anti-allergic effects of polyphenols such as lotus leaf polyphenol extract(LPE)and Polygonatum polyphenol extract(PPE),MLP was extracted to construct a zebrafish food allergy model.Zebrafish were simultaneously administered LPE and PPE.The allergenic effects of MLP and the anti-allergic effects of LPE and PPE were investigated by behavioral analysis,intestinal section staining,and real time fluorescent quantitative polymerase chain reaction(qRT-PCR).The results indicated that MLP reduced the locomotor activity of zebrafish,while LPE and PPE could restore this indicator,demonstrating a protective effect.MLP caused significant damage to the intestinal barrier,whereas LPE and PPE enhanced immune responses,promoted intestinal recovery and alleviated allergic symptoms.qRT-PCR result showed that LPE and PPE regulated the expression of inflammatory factors such as interleukin-6(IL-6),interleukin-1β(IL-1β),interleukin-4/13B(IL-4/13B),and interleukin-10(IL-10),thereby inhibiting the inflammatory response induced by MLP.This study confirms the allergenicity of MLP and the anti-allergic potential of LPE and PPE,providing a scientific basis for the prevention and treatment of food allergies.
