表里井穴刺血法对脂多糖致热家兔前列腺素E2水平的影响OA
Effects of Bloodletting Therapy at Exterior-Interior Jing Acupoints on Prostaglandin E2 Levels in Rabbits with LPS-Induced Fever
目的 探寻表里井穴刺血法的退热机制.方法 将30只雄性家兔随机分为正常组、模型组、少商组、商阳组和少商商阳组,通过静脉注射脂多糖构建外感发热模型.造模成功后分别于双侧少商穴、商阳穴、少商加商阳穴刺血,通过分析干预后各组家兔血清和脑组织中前列腺素E2(PGE2)、环氧化酶-2(COX-2)水平,探索退热机制.结果 与模型组相比,少商组、商阳组、少商商阳组家兔肛温降低差异均有统计学意义(P<0.01),与正常组比较,少商组、商阳组、少商商阳组家兔肛温均无明显差异(P>0.05).与模型组比较,少商组、少商商阳组家兔血清PGE2水平明显降低(P<0.05或P<0.01),商阳组、少商商阳组家兔脑组织PGE2水平明显降低(P<0.05或P<0.01),少商组、商阳组、少商商阳组家兔下丘脑COX-2水平降低,差异有统计学意义(P<0.05或P<0.01).结论 少商、商阳穴刺血可能通过抑制COX-2/PGE2信号通路,减少中枢及外周致热介质释放发挥退热作用.
Objective:To explore the antipyretic mechanism of bloodletting therapy at exterior-interior Jing acu-points.Methods:30 male rabbits were randomly divided into normal group,model group,Shaoshang group,Shang-yang group and Shaoshang-Shangyang group.An exogenous fever model was established by intravenous injection of lipopolysaccharide(LPS).After successful modeling,bloodletting therapy was performed at bilateral Shaoshang acupoint,bilateral Shangyang acupoint,and bilateral Shaoshang plus Shangyang acupoint respectively.The anti-pyretic mechanism was explored by analyzing the levels of prostaglandin E2(PGE2)and cyclooxygenase-2(COX-2)in serum and brain tissue of rabbits in each group after intervention.Results:Compared with the model group,the anal temperature of rabbits in the Shaoshang group,Shangyang group and Shaoshang-Shangyang group de-creased with statistical significance(P<0.01).Compared with the normal group,there was no statistical difference in the anal temperature of rabbits in the Shaoshang group,Shangyang group and Shaoshang-Shangyang group(P>0.05).Compared with the model group,the serum PGE2 level of rabbits in the Shaoshang group decreased with statistical significance(P<0.05),and that in the Shaoshang-Shangyang group decreased with significant statistical significance(P<0.01).The brain tissue PGE2 level of rabbits in the Shangyang group decreased with significant statistical significance(P<0.01),and that in the Shaoshang-Shangyang group decreased with statistical significance(P<0.05).The hypothalamic COX-2 level of rabbits in the Shaoshang group decreased with significant statistical significance(P<0.01),and that in the Shangyang group and Shaoshang-Shangyang group decreased with statisti-cal significance(P<0.05).Conclusion:Bloodletting therapy at Shaoshang and Shangyang acupoint may exert an antipyretic effect by inhibiting the COX-2/PGE2 signaling pathway and reducing the release of central and periph-eral pyrogenic mediators.
