2024-2025年天津市GⅡc亚型猪流行性腹泻病毒S基因遗传变异分析OA
Genetic Variation Analysis on S Gene of GⅡc Subtype PEDV in Tianjin City in 2024-2025
为探究2024年以来天津市猪流行性腹泻病毒(PEDV)的遗传变异特征,对采自天津市5个区县27个中小型猪场的127份临床样品进行RT-qPCR检测,选择代表性阳性样品进行S基因扩增测序及系统发育分析.结果显示:在127份临床样品中,共检出46份阳性样品.在18份代表性阳性样品中,成功获得18条S基因全长序列;系统发育分析表明,所有18条序列均属于GⅡc亚型,且与经典疫苗株CV777的同源性仅为92.2%~93.5%.氨基酸序列分析发现,所有序列在55~56 aa(插入IGEN)、135~136 aa(插入D/N)及155~156 aa(缺失DI)位点,均呈现GⅡ型变异株的典型分子特征;与GⅡb型疫苗株AJ1102相比,S1区氨基酸突变率高达50.0%~73.5%,关键突变位点位于NTD和COE等重要抗原结构域;N-糖基化分析显示,阳性序列的糖基化模式与GⅡ型高毒力株高度一致,特别是在57ENQGVNST64和722NSTF725区域.结果表明:GⅡc亚型PEDV已成为天津市优势流行毒株,其S基因的持续变异可能导致病毒抗原性改变以及免疫逃逸能力增强.本研究结果为PED疫苗研发与防控策略制定提供了重要依据.
In order to investigate the genetic variation characteristics of porcine epidemic diarrhea virus(PEDV)in Tianjin City since 2024,127 clinical samples collected from 27 small-to-medium swine farms across 5 districts/counties of Tianjin City were tested using RT-qPCR,from which representative positive samples were selected for S gene amplification,sequencing and phylogenetic analysis.The results showed 46/127 positive samples were detected.18 full-length S gene sequences were successfully obtained from 18 representative positive samples.Phylogenetic analysis indicated that all the 18 sequences belonged to GⅡc subtype,92.2%-93.5%identical to CV777,the classical vaccine strain.Amino acid sequence analysis revealed that typical molecular features of GⅡ-type variants appeared at positions 55-56 aa(IGEN insertion),135-136 aa(D/N insertion)and 155-156 aa(DI deletion)in all the sequences.Compared to AJ1102,the GⅡb vaccine strain,the mutation rate of amino acid in the S1 region was up to 50.0%-73.5%,with key mutation sites located in critical antigenic domains such as N-terminal domain(NTD)and coronavirus epitope(COE).N-glycosylation analysis revealed that the glycosylation patterns of positive sequences were highly consistent with those of highly pathogenic GⅡ strains,particularly in the 57ENQGVNST64 and 722NSTF725 regions.In conclusion,GⅡc subtype PEDV had been dominant in Tianjin City,and changes of virus antigenicity and enhanced capabilities of immune escape could be caused by the continuous mutations of its S gene.Key evidences were provided by the study for development of PED vaccines and formulation of control strategies.
