重组牛冠状病毒N蛋白在昆虫杆状病毒表达系统中的表达与鉴定OA
Expression and Identification of Recombinant Bovine Coronavirus N Protein in Insect Baculovirus Expression Vector System
为更好利用昆虫杆状病毒表达系统对牛冠状病毒(BcoV)的主要结构蛋白N蛋白进行表达,本研究采用BCoV-N基因为目标设计特异性引物进行PCR扩增,通过无缝克隆技术将其插入昆虫杆状病毒系统转移载体pFastBacHT B,并构建pFastBacHT B-BCoV-N重组质粒.随即转化至DH10Bac大肠杆菌,经过蓝白斑筛选后提取阳性重组杆粒Bacmid-BCoV-N,转染至Sf9昆虫细胞产生杆状病毒并进行传代.结果表明,经双酶切、PCR验证,重组质粒与重组杆粒均构建成功,重组杆粒Bacmid-BCoV-N转染Sf9昆虫细胞并连续盲传5代后获得重组杆状病毒Bacmid-BCoV-N,成功表达纯化出BCoV-N蛋白,经SDS-PAGE及Western blot鉴定,其分子量大小约为55 kDa,具有良好的特异性.将50 μg重组蛋白与弗氏完全佐剂充分混匀乳化后免疫小鼠,间接ELISA试验结果显示免疫小鼠产生了特异性抗体.综上所述,本研究构建的昆虫杆状病毒表达系统能够高水平地表达外源基因,可以满足后续建立免疫学抗体诊断检测技术的需要.
In this study,the insect baculovirus expression system was used to express N protein,amplify the BCoV-N gene by PCR and inserted into the insect baculovirus system transfer vector pFastBacHT B by seamless cloning to con-struct the pFastBacHT B-BCoV-N recombinant plasmid.After transforming into DH10Bac E.coli,positive recombi-nant Bacmid-BCoV-N was extracted through blue-white spot screening,and transfected into Sf9 insect cells to pro-duce baculovirus and then transited.The results showed that both validation the recombinant plasmid and the recom-binant bacmid were successfully constructed.Following transfection of Sf9 insect cells with the recombinant bacmid Bacmid-BCoV-N and five successive blind passages,recombinant baculovirus Bacmid-BCoV-N was successfully obtained,enabling the expression and purification of BCoV-N protein,and SDS-PAGE and Western blot analysis identified a molecular weight of approximately 55 kDa,demonstrating good specificity.For immunological evaluation,50 μg of the recombinant protein emulsified with Freund's complete adjuvant was administered intramuscularly to mice.Indirect ELISA results confirmed the induction of specific antibodies in immunized mice.In conclusion,the in-sect baculovirus expression system established in this study enables high-level expression of exogenous genes,ful-filling the requirements for subsequent development of immunological diagnostic assays for antibody detection.
