本研究旨在克隆多浪羊促甲状腺激素释放激素受体基因(Thyrotropin-releasing hormone receptor,TRHR),并检测与繁殖相关的组织中TRHR基因的表达情况,为探求其在绵羊繁殖中的作用提供理论基础.试验以多浪羊初情期垂体cDNA为模板,通过RT-PCR扩增TRHR基因,将所得产物克隆测序;利用生物信息学软件预测多浪羊TRHR蛋白结构和理化性质;利用qPCR技术检测多浪羊初情期前后3个时期5个繁殖相关组织中TRHR基因的表达水平.结果表明,TRHR基因克隆所得序列大小为1 373 bp,其中CDS区为1 197 bp,与GenBank上预测的绵羊mRNA序列相似性达99.83%;通过系统进化树分析发现,多浪羊TRHR基因与羚羊的遗传距离最近,而与北美水獭的遗传距离最远;qPCR试验结果表明,多浪羊TRHR基因在不同时期、不同组织中均有显著表达,其中在初情期阶段垂体中的表达量显著高于其他组织(P<0.05).综上,本研究成功克隆了多浪羊TRHR基因编码区序列,并发现该基因在多浪羊初情期垂体中呈现高表达特征.

The aim of this study was to clone the TRHR(Thyrotropin-releasing hormone receptor)gene of Duolang sheep and to detect the expression of the TRHR gene in the tissues related to reproduction,so as to provide a theoretical basis for exploring the role of the TRHR gene in sheep reproduction.The cDNA of puberty pituitary gland of Duolang sheep was used as template for the experiment,and the TRHR gene was amplified by RT-PCR and the resulting product was cloned and sequenced.Bioinformatics software was used to predict the structure and physicochemical properties of TRHR protein in Duolang sheep.The expression levels of TRHR gene in five tissues related to reproduction in three pe-riods before and after puberty in Duolang sheep were also detected by qPCR.The results showed that the sequence size of the cloned TRHR gene was 1 373 bp,of which the CDS region was 1 197 bp,with 99.83%similarity to the mRNA se-quence of sheep predicted on GenBank.Phylogenetic tree analysis revealed that the TRHR gene of Duolang sheep had the closest genetic distance to Oryx dammah and the furthest to Lontra canadensis.qPCR assay showed that the TRHR gene of Duolang sheep was significantly expressed in different periods and tissues,and at the puberty stage,the expres-sion of the TRHR gene in the pubertal pituitary gland was significantly higher than that in other tissues(P<0.05).In sumamry,the present study successfully cloned the sequence of the coding region of TRHR gene in Duolang sheep and found that the gene showed high expression characteristics in the pubertal pituitary gland of Duolang sheep.
