本试验旨在探究在circCOL1A1过表达及敲减的毛囊干细胞中差异表达的基因,初步筛选调控长江三角洲白山羊毛囊干细胞生长发育的关键基因.对分离自115~125d胎龄的长江三角洲白山羊公羔颈脊部皮肤组织的毛囊干细胞进行circCOL1A1的过表达和敲减处理,将处理后的细胞分别进行转录组测序和生物信息学分析,利用STRING等数据库构建多个相互作用网络.通过实时定量聚合酶链反应和Sanger测序进一步验证差异表达基因.结果表明,MYO16、HAPLN4、TBKBP1、CPXM1、TXINP、CSRP2、THEM5基因在 circCOL1A1 敲减的毛囊干细胞中差异表达,CMTM3、STAT1、MX1、SAMD9、GBP5等基因在circCOL1A1过表达的毛囊干细胞中差异表达;这些基因通过PI3K-Akt、RIG-Ⅰ样受体(RLR)、JAK-STAT和MAPK-Ras-ERK等信号通路参与毛囊干细胞的生长发育.说明MYO16、HAPLN4、CMTM3、STAT1等差异表达基因主要富集在PI3K-Akt、RLR等通路中,参与调控毛囊干细胞的生长发育等过程.

This study aimed to investigate the differentially expressed genes(DEGs)in hair follicle stem cells(HFSCs)with circCOL1A1 overexpression or knockdown,and to preliminarily screen key genes regulating HFSCs growth and development in Yangtze River Delta White goat.HFSCs were isolated from the cervical spinal skin tissues of male Yangtze River Delta White Goat fetuses at 115-125 d of gestation.After overexpression or knockdown of circ-COL1A1,transcriptome sequencing and bioinformatics analysis were performed on the treated cells,followed by mul-tiple interaction networks that constructed by STRING and other databases.Finally,DEGs were further validated using real-time quantitative polymerase chain reaction(qPCR)and Sanger sequencing.The results showed that genes such as MYO16,HAPLN4,TBKBP1,CPXM1,TXINP CSRP2,and THEM5 were differentially expressed in HFSCs with knock-down of circCOL1A1,while genes including CMTM3,STAT1,MX1,SAMD9,and GBP5 were differentially expressed in HFSCs with overexpression of circCOL1A1.These genes participated in HFSCs growth and development through PI3K-Akt,RIG-Ⅰ-like receptor(RLR),JAK-STAT,and MAPK-Ras-ERK signaling pathways.This study found that differentially expressed genes such as MYO 16,HAPLN4,CMTM3,and STAT1 were mainly enriched in the PI3K-Akt and RLR pathways,and involved in regulating processes of growth and development of hair follicle stem cells.
