本研究通过比较藏羊、杜泊羊(♂)×藏羊(♀)杂交一代羊(杜藏F1羊)与杂交育成代(海东黑头羊)的高原低氧适应性能,旨在为高海拔地区绵羊新品种的选育提供理论基础.选取12月龄藏羊、杜藏F1羊与海东黑头羊公羊各20只,采集静脉血检测血液生理指标;每组随机选择6只,称重后进行屠宰试验,测定心、肺的脏器系数及其结构特征;随后通过转录组测序分析3组羊肺组织的基因表达模式.结果表明,杜藏F1羊的血液红细胞数和血红蛋白浓度均显著低于藏羊(P<0.05),而海东黑头羊与藏羊的各项血液生理指标均无显著差异(P>0.05).3组羊的心脏重量无显著差异(P>0.05),组织形态亦无明显区别(P>0.05).与藏羊相比,杜藏F1羊的肺泡面积显著减少(P<0.05),肺泡壁厚度显著增加(P<0.05),而海东黑头羊的肺泡面积显著高于杜藏F1羊(P<0.05),肺泡壁厚度则显著低于杜藏F1羊(P<0.05).肺组织的转录组分析结果表明,藏羊与杜藏F1羊间有474个差异表达基因,而海东黑头羊与杜藏F1羊间有1 752个差异表达基因.基因功能富集显示,相比于藏羊与海东黑头羊,杜藏F1羊上调的基因涉及血管生成、组织纤维化等通路,而特异性下调基因则富集于免疫应答、应激响应等通路.综上所述,杜藏F1羊的肺脏组织发育与功能存在一定缺陷,而海东黑头羊在遗传上则融合了藏羊的低氧适应性与杜泊羊的优势生产性能.

This study aimed to compare the high-altitude hypoxic adaptation among Tibetan sheep,Dorper sheep(♂)× Tibetan sheep(♀)F1 sheep(referred to as DT F1)and Haidong Black-headed sheep(hybrid breeding generation),thereby providing a theoretical foundation for new breeding sheep in high-altitude regions.Twenty male 12-month-old sheep were selected from each group for blood sample collection to assess hematological parameters.Six sheep were randomly selected from each group,weighed,and then subjected to slaughter experiment to determine the heart and lung coefficients and their morphological characteristics.Subsequently,transcriptome analysis was performed to compare gene expression patterns in lung tissues among the three groups.The results showed that the red blood cell count and hemoglobin concentration of DT F1 were significantly lower than those of Tibetan sheep(P<0.05),while there were no significant differences in all hematological parameters between Haidong Black-headed sheep and Ti-betan sheep(P>0.05).No significant difference was observed in heart weight among the three groups(P>0.05).No sig-nificant difference was also observed in tissue morphology among the groups(P>0.05).However,compared to Tibetan sheep,DT F1 exhibited significant reduced in alveolar area and increased in alveolar wall thickness(P<0.05).The alveo-lar area of Haidong Black-headed sheep showed a significant increase(P<0.05),and alveolar wall thickness showed a significant reduced compared with that of DT F1(P<0.05).Transcriptome analysis of lung tissues revealed 474 differen-tially expressed genes between DT F,and Tibetan sheep,and 1 752 differentially expressed genes between Haidong Black-headed sheep and DT F1.Functional enrichment analysis indicated that genes upregulated in DT F,(compared to Tibetan and Haidong Black-headed sheep)were associated with pathways such as angiogenesis and tissue fibrosis,whereas the specifically downregulated genes were enriched in immune response and stress response pathways.In con-clusion,the lung tissue development and function of DT F1 exhibited certain deficiencies,whereas the Haidong Black-headed sheep integrates the hypoxic adaptability of Tibetan sheep and the superior production performance of Dorper sheep.
