卷曲乳杆菌及徳氏乳杆菌上清液对阴道加德纳菌及大肠埃希菌的作用OA
Study on the effects of Lactobacillus crispatus and Lactobacillus delbrueckii supernatants on Gardnerella Vaginalis and Escherichia Coli
目的:探究卷曲乳杆菌及徳氏乳杆菌上清液及二者混合对阴道加德纳菌及大肠埃希菌的菌量和生物膜的抑制作用.方法:选取从阴道分泌物中分离出的 5 株卷曲乳杆菌及分离自阴道用乳杆菌活菌胶囊的徳氏乳杆菌,通过结晶紫染色法测定生物膜形成能力.制备卷曲乳杆菌、徳氏乳杆菌及二者混合培养上清液,与阴道加德纳菌及大肠埃希菌共培养.测量OD值,结晶紫染色法及Live/Dead染色后激光共聚焦显微镜分别测量上述乳杆菌对阴道加德纳菌及大肠埃希菌菌量、生物膜形成能力及生物膜厚度的作用.结果:5 株卷曲乳杆菌及徳氏乳杆菌均于培养48h的生物膜形成能力最强(P<0.05).卷曲乳杆菌对阴道加德纳菌生长无明显抑制作用,但对其生物膜形成有较强的抑制作用(P<0.01);对大肠埃希菌无生长抑制作用,可促进大肠埃希菌生物膜的形成(P<0.05).德氏乳杆菌可抑制阴道加德纳菌及大肠埃希菌的生长(P<0.01),对大肠埃希菌的抑制作用强于卷曲乳杆菌(P<0.05);同时可抑制阴道加德纳菌生物膜的形成(P<0.05),但对大肠埃希菌生物膜的形成有促进作用.卷曲乳杆菌及德氏乳杆菌间存在协同作用,可加强对大肠埃希菌的生长和生物膜抑制作用.结论:卷曲乳杆菌对阴道加德纳菌的生物膜形成有较强抑制作用,但对大肠埃希菌无明显抑制作用.徳氏乳杆菌可抑制阴道加德纳菌的生长和生物膜形成.卷曲乳杆菌和徳氏乳杆菌间存在协同作用.
Objective:To investigate the inhibitory effects of Lactobacillus crispatus(L.crispatus)supernatant,Lactobacillus delbrueckii(L.delbrueckii)supernatant,and their mixture on the bacterial counts and biofilms of Gardnerella Vaginalis(G.vaginalis)and Escherichia Co-li(E.coli).Methods:Five strains of L.crispatus isolated from vaginal secretions and L.del-brueckii isolated from Live Lactobacillus Capsule for Vaginal Use were selected.Biofilm forming capacity was determined using the Crystal Violet(CV)method.The supernatants from the cul-tures of L.crispatus,L.delbrueckii,and their mixture were prepared and co-cultured with G.vag-inalis and E.coli.The effects of these Lactobacillus on the bacterial counts,biofilm forming ca-pacity,and biofilm thickness of G.vaginalis and E.coli were measured using OD values,CV method,and Live/Dead staining followed by confocal laser scanning microscopy.Results:All five strains of L.crispatus and L.delbrueckii exhibited the strongest biofilm forming ability after 48 hours(P<0.05).L.crispatus had no significant inhibitory effect on the growth of G.vaginalis but strongly inhibited its biofilm formation(P<0.01).It did not inhibit the growth of E.coli but promoted the formation of its biofilm.L.delbrueckii inhibited the growth of G.vaginalis and E.coli(P<0.01),with a stronger inhibitory effect on E.coli than L.crispatus(P<0.05).It al-so inhibited the biofilm formation of G.vaginalis(P<0.05)but promoted the biofilm formation of E.coli.There is a synergistic effect between L.crispatus and L.delbrueckii,which enhances the growth and biofilm formation of E.coli.Conclusion:L.crispatus has a strong inhibitory effect on biofilm formation by G.vaginalis but no significant inhibitory effect on E.coli.L.delbrueckii can inhibit the growth and biofilm formation of G.vaginalis.There is a synergistic effect between L.crispatus and L.delbrueckii.
