Screening of physiological indicators for winter desiccation resistance and study on winemaking charac-teristics of Hybrid wine grape selection'140902'
[目的]本研究旨在探究优系'140902'葡萄枝条的抗抽干生理特性及其酿酒品质,以验证其作为优质酿酒葡萄在干旱低温地区推广的可行性.[方法]以'北红'和抗抽干新品系'140902'一年生枝条为材料,在自然越冬下,研究其生理生化指标的差异,综合评价葡萄抗抽干性差异,并筛选相关指标.基于变异检测,通过KEGG富集分析筛选抗抽干有关候选基因,利用实时荧光定量分析和相关基因酶活性测定探究其在葡萄抗抽干中发挥的作用.同时,结合HS-SPME-GC-MS测定2种基因型葡萄果实香气等品质指标并进行葡萄酒感官分析,以探究其酿酒特性.[结果]自然越冬条件下,'140902'的抽干率显著低于'北红',且葡萄一年生枝条抽干率与总果胶、纤维素含量等7个指标显著相关.通过主成分分析得到4个主成分,可反映原耐抽干指标信息的92.04%;通过隶属函数分析可见,'140902'和'北红'抗抽干性综合评价平均D值分别为0.641和0.534.KEGG富集分析获得3个与抽干相关的候选基因,其在2个品种(系)中的表达模式存在显著差异.对葡萄浆果品质研究发现'140902'的单宁、总酚、总酸含量等6个指标均显著高于'北红',且香气以青香和果香为主.[结论]总果胶含量、过氧化氢酶活性、过氧化物酶活性等7个指标可作为判断抗抽干的主要指标.与'北红'相比,'140902'的抗抽能力更强,且'140902'的葡萄酒典型性强,是可在干旱和半干旱区自然越冬的优质酿酒葡萄新品系.
[Objective]This study aimed to explore the physiological characteristics of winter desiccation resistance and the eno-logical quality of the superior grape line'140902',verifying its feasibility for promotion as a high-quality wine grape in arid and low-temperature areas.[Methods]Using one-year-old shoots of'Beihong'and the desiccation-resistant selection'140902'as materials,physiological and biochemical indicators were studied under natural overwintering conditions to comprehensively evaluate and screen for resistance markers.Based on variation detection,candidate genes related to desiccation resistance were identified via KEGG enrichment analysis.Their roles were further explored through real-time fluorescence quantitative(RT-qPCR)analysis and enzyme activity assays.Meanwhile,berry aroma and quality traits were determined using HS-SPME-GC-MS,followed by sensory analysis to evaluate winemaking characteristics.[Results]Under natural overwintering,the desicca-tion rate of'140902'was significantly lower than that of'Beihong'.The shoot desiccation rate correlated significantly with sev-en indicators,including total pectin and cellulose content.Principal Component Analysis extracted four principal components,representing 92.04%of the original data.Membership function analysis showed that the average comprehensive evaluation D-values for'140902'and'Beihong'were 0.641 and 0.534,respectively.KEGG analysis identified three desiccation-related can-didate genes with significantly different expression patterns between the two genotypes.Furthermore,'140902'berries exhibit-ed significantly higher tannins,total phenols,and total acid content than'Beihong',with an aroma profile dominated by green and fruity notes.[Conclusion]Seven indicators,including total pectin,CAT,and POD activity,can serve as key indicators for assessing desiccation resistance.Compared to'Beihong','140902'exhibited superior desiccation resistance and strong wine typicity,making it a promising high-quality selection for arid and semi-arid regions.
