小偃麦衍生系抗旱基因功能分子标记检测及效应分析OA
Detection and Effect Analysis of Functional Molecular Markers of Drought-Resistant Genes in Wheat-Thinopyrum intermedium Introgression Lines
为进一步明确小偃麦衍生系抗旱等位基因组成,采用5个小麦抗旱基因TaNRX-B1、TaDreb-B1、Ta-PYL1-1B、TaBADH-A1和TaDRO-5B的功能分子标记检测了175份小偃麦衍生系,并结合萌发期13个抗旱指标的抗旱系数和综合评价D值,分析了各等位基因的抗旱效应.结果显示,5个抗旱等位基因在小偃麦衍生系中的频率总体偏低,其中,TaDreb-B1a的频率最高,为38.86%;TaDRO-5B基因的Hap-5B-I的频率最低,为 5.14%.在 117份中等以上抗旱性衍生系中,2份携带 4个抗旱等位基因的衍生系CH6310(TaDreb-B1a/TaBADH-A1b/TaPYL1-1BIn-442/Hap-5B-I)和 CH1539(TaNRX-B1a/TaDreb-B1a/TaBADH-A1b/Hap-5B-I)均为强抗旱种质;13份强抗旱衍生系未检测到任何抗旱等位基因,可能存在未知的抗旱相关基因.携带抗旱等位基因TaDreb-B1a、TaPYL1-1BIn-442和Hap-5B-I种质的多数指标抗旱系数均值高于携带其干旱敏感等位基因的衍生系,且抗旱等位基因与多个指标抗旱系数呈显著正相关,抗旱效应较高.因此,抗旱基因TaDreb-B1、TaPYL1-1B和TaDRO-5B的功能标记能够有效用于小偃麦衍生系抗旱基因型检测及分子标记辅助选择育种.
In order to further clarify the favor alleles of drought-resistant gene composition in the wheat-Thinopyrum intermedium introgression lines,in this study,the functional molecular markers of 5 wheat drought resistant genes,including TaNRX-B1,TaDreb-B1,TaPYL1-1B,TaBADH-A1,and TaDRO-5B,were used to detect 175 wheat-Thinopyrum intermedium introgression lines.The drought resistance effect of each allele was analyzed by combining the drought resistance coefficients of 13 drought-resistant indicators and the comprehensive evaluation D value at the germination stage.The results showed that the frequency of the 5 drought-resistant alleles was generally low in the introgression lines,among which TaDreb-B1a had the highest proportion(38.86%)and Hap-5B-I allele of TaDRO-5B gene had the lowest proportion(5.14%).Among the 117 lines with moderate or above drought resistance,CH6310(TaDreb-B1a/TaBADH-A1b/TaPYL1-1BIn-442/Hap-5B-I)and CH1539(TaNRX-B1a/TaDreb-B1a/TaBADH-A1b/Hap-5B-I),carrying 4 drought-resistant alleles,were highly drought-resistant lines.As the five drought-resistant alleles were not detected in the 13 highly drought-resistant lines,there might be new unknown drought-resistant genes.The average drought resistance coefficients of most indicators of introgression lines carrying drought-resistant alleles TaDreb-B1a,TaPYL1-1BIn-442 and Hap-5B-I were higher than those of introgression lines carrying their drought-sensitive alleles.The drought-resistant alleles were significantly positively correlated with the drought resistance coefficients of multiple indicators,and the drought resistance effect was higher.Therefore,the functional molecular markers of drought-resistant genes such as TaDreb-B1,TaPYL1-1B and TaDRO-5B could be effectively used for drought-resistant genotype detection and molecular marker-assisted selection breeding in wheat-Thinopyrum intermedium introgression lines.
