舒润修护霜对皮肤损伤修护的作用研究OA
Study on the repairing effects of a moisturizing repair cream on skin damage
研究含有白藜芦醇、和厚朴酚、七叶皂苷的舒润修护霜对皮肤损伤的修复作用.采用体外生化实验检测修护霜对自由基的清除效果;通过构建细胞紫外光急性损伤模型,评估其对HaCaT细胞中氧化应激损伤的影响;采用细胞划痕实验评估其对皮肤细胞修护的作用;使用DNFB诱导小鼠特异性皮炎模型,观察小鼠皮损程度,记录搔抓次数,分析皮损组织病理学变化,并通过实时荧光定量聚合酶链式反应(RT-qPCR)法检测皮损组织中相关细胞因子mRNA的表达水平.由实验结果可知,修护霜对自由基ABTS及DPPH均具有较强的清除活性;显著抑制紫外线诱导的Hacat细胞中活性氧(ROS)的生成,并增强细胞活力;促进细胞增殖及皮肤细胞损伤修复;在特应性皮炎小鼠模型中,修护霜组的皮损程度、搔抓次数、表皮增厚及炎性细胞浸润均显著减少;修护霜组中IL-1β、TNF-α、IL-6、IL-4、IL-13的mRNA表达显著低于模型组.舒润修护霜可通过抑制皮肤细胞的氧化应激损伤及炎症反应,促进皮肤细胞增殖,从而发挥皮肤损伤修护功效.
This study evaluates the repairing effects of a moisturizing repair cream formulated with resveratrol,magnolol,and aescin on skin damage.In vitro biochemical assays were used to determine the cream's free radical-scavenging capacity.An acute UV-induced HaCaT cell injury model was established to assess its protective effects against oxidative stress.A cell scratch assay was employed to examine its role in promoting skin cell repair.A DNFB-induced mouse model of contact dermatitis was used to evaluate improvements in skin lesion severity,record scratching frequency,analyze histopathological changes in the affected tissue,and quantify mRNA expression levels of key cytokines via real-time quantitative PCR.The results showed that the repair cream exhibited strong ABTS and DPPH free radical-scavenging activity;significantly inhibited reactive oxygen species(ROS)generation in UV-exposed HaCaT cells and enhanced cell viability;and promoted cell proliferation and repair of damaged skin cells.In the atopic-dermatitis-like mouse model,the repair cream notably reduced skin lesion severity,scratching frequency,epidermal thickening,and inflammatory cell infiltration.Additionally,the mRNA expression levels of IL-1β,TNF-α,IL-6,IL-4,and IL-13 in the treated group were significantly lower than those in the model group.Overall,the moisturizing repair cream exerts its skin-repairing effects by suppressing oxidative stress and inflammatory responses while promoting skin cell proliferation.
