A Comparative Study on the Tetraploid Induction Methods of Allotetraploid Oyster by Cytochalasin B and Hypotonic Treatment

为探索细胞松弛素B(Cytochalasin B,CB)和低渗抑制第一极体诱导GGAG(GGA ♀×GG♂)和AAGG(AAG ♀×GG♂)两种异源四倍体的最佳条件,以卵裂率、孵化率、四倍体率及诱导效率为指标,研究了不同条件(CB浓度:0.2、0.4、0.6和0.8 mg/L,盐度:6、8、10和12,诱导持续时间:10、15、20和25 min)对两种异源四倍体(GGAG和AAGG)诱导效果的影响,并分析了早期幼虫的生长和存活情况.研究表明,采用0.6 mg/L CB,持续处理时间15 min诱导GGAG,幼虫四倍体率为(42.75±2.89)%;采用盐度8,持续处理时间15 min诱导GGAG,幼虫四倍体率为(32.28±1.97)%.采用0.6 mg/L CB,持续处理时间20 min诱导AAGG,幼虫四倍体率为(47.03±2.66)%;采用盐度10,持续处理时间15 min诱导AAGG,幼虫四倍体率为(33.06±1.18)%.

To investigate the tetraploid induction effect between two allotetraploid oysters(GGAG—GGA ♀×GG ♂ and AAGG—AAG ♀×GG ♂)under varying cytochalasin B(CB)concentration(0.2,0.4,0.6,0.8 mg/L),salinity gradient(6,8,10,12)and induction duration(10,15,20,25 min),we analyzed the cleavage rate,hatching rate,tetraploidy rate and induction efficiency.At the same time,the growth and survival of the early larvae were analyzed.The results showed that the optimal method of inducing tetraploid of GGAG was 0.6 mg/L of CB and 15 min.The tetraploidy rate of(42.75±2.89)%was obtained with the method.The salinity 8 and 15 min was also the optimal method,with which the tetraploidy rate of(32.28±1.97)%was obtained.For AAGG,0.6 mg/L of CB and 20 min was optimal,with which the tetraploidy rate of(47.03±2.66)%was obtained.The salinity 10 and 15 min was the optimal method,with which the tetraploidy rate of(33.06±1.18)%was obtained.
