基于PI3K/AKT通路探讨中医药治疗糖脂代谢性疾病OA
Exploring Traditional Chinese Medicine in the Treatment of Glycolipid Metabolic Diseases Based on the PI3K/AKT Pathway
代谢性疾病是由遗传、环境等多种因素共同作用而导致的慢性疾病,其发生与进展常伴随着机体内糖、脂、蛋白质等多种物质代谢的失衡或紊乱.磷脂酰肌醇-3-激酶/蛋白激酶 B(phosphatidyl-inositol 3-kinase/serine-threonine kinase,PI3K/AKT)信号通路是调控机体新陈代谢的核心通路之一,在多种代谢性疾病的发生与发展中发挥关键调控作用.中医药可通过调节PI3K/AKT信号通路,影响细胞的增殖、分化、凋亡及自噬等过程,进而改善糖、脂、蛋白质等代谢紊乱状态,在代谢性疾病的防治中展现出重要作用.然而,代谢性疾病的发病机制涉及多条信号通路、多个分子靶点,它们通过复杂的调控网络共同影响疾病进程.目前多数研究仍集中于单方、单药对单一细胞中某条通路或少数靶点的作用,缺乏系统性与深入性的整体机制探索.因此,未来需进一步揭示PI3K/AKT信号通路与其他细胞、分子及通路之间的交互关系,阐明中医药在多靶点、多通路层面的整体调节机制,从而为代谢性疾病的预防与治疗提供新策略,也为相关药物的研发与优化提供新思路.
Metabolic diseases are chronic conditions resulting from the combined effects of genetic,environmental,and other factors,and have become a major global health threat.The PI3K/AKT signaling pathway is one of the core pathways regulating body metabolism and plays a crucial regulatory role in the occurrence and development of various metabolic diseases.The onset and progression of this kind of diseases are often accompanied by imbalances or disorders in the metabolism of various substances within the body,such as sugars,lip-ids,and proteins.TCM can influence processes like cell proliferation,differentiation,apoptosis,and autophagy,etc.by modulating the PI3K/AKT signaling pathway,thereby improving metabolic disturbances of glucose,lipids,proteins,etc.,and demonstrating significant potential in the prevention and treatment of metabolic diseases.However,the pathogenesis of metabolic diseases involves multiple signa-ling pathways and molecular targets,which collectively affect disease progression through complex regulatory networks.Most current re-search remains still focuses on the effects of single formula or medicine on a specific pathway or a limited number of targets within a sin-gle cell type,lacking systematic and in-depth exploration of the holistic mechanisms.Therefore,it is necessary to further elucidate the interactions between the PI3K/AKT signaling pathway and other cells,molecules,and pathways in the future,and clarify the holistic reg-ulatory mechanisms of TCM at the multi-target and multi-pathway levels,which will provide new strategies for the prevention and treat-ment of metabolic diseases and offer new insights for the development and optimization of related medicines.
