消囊调经汤调控NF-κB/IRS-1/PI3K/AKT通路治疗肥胖型多囊卵巢综合征的作用机制OA
Mechanism of Xiaonang Tiaojing Decoction in Treating Obese Polycystic Ovary Syndrome by Regulating the NF-κB/IRS-1/PI3K/AKT Pathway
目的:探讨消囊调经汤通过调控核因子-κB(nuclear factor kappa-B,NF-κB)/胰岛素受体底物-1(insulin receptor sub-strate-1,IRS-1)/磷脂酰肌醇3 激酶(phosphatidylinositol-3-kinase,PI3K)/蛋白激酶B(protein kinase B,AKT)通路治疗肥胖型多囊卵巢综合征(polycystic ovary syndrome,PCOS)的作用机制.方法:采用高脂饮食联合脱氢表雄酮构建肥胖型PCOS小鼠模型,分为正常对照组(Control组)、模型组(Model组)、二甲双胍组(200 mg·kg-1,Met组)、消囊调经汤低剂量组(5g·kg-1,XNTJT-L)及消囊调经汤高剂量组(20g·kg-1,XNTJT-H),每组10 只,灌胃8 周.卵巢颗粒细胞分为空白组(Control组)、模型组(Model组)、含药血清组(XNTJT组)、含药血清+si-NC组(XNTJT+si-NC组)及含药血清+si-IRS-1 组(XNTJT+si-IRS-1 组).检测小鼠胰岛素抵抗指数(HOMA-IR)与血清游离脂肪酸(free fatty acids,FFA)水平,HE染色检测小鼠动情周期与卵巢组织病理形态,ELISA检测小鼠血清黄体生成素(luteinizing hormone,LH)、卵泡刺激素(follicle-stimulating hor-mone,FSH)、睾酮(testosterone,T)及LH/FSH水平,Western blot检测小鼠和细胞NF-κB/IRS-1/PI3K/AKT通路关键蛋白表达水平,ELISA检测小鼠和细胞肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)及NF-κB水平,CCK-8 法检测细胞增殖率,流式细胞术检测细胞凋亡率.结果:动物实验:与Control组比较,Model组小鼠HOMA-IR和FFA水平升高(P<0.01).与 Model 组比较,Met 组和 XNTJT-H 组小鼠 HOMA-IR、FFA 水平降低(P<0.05).Control组小鼠动情周期规律,可见不同发育阶段的卵泡.Model组小鼠动情周期紊乱,初级卵泡、次级卵泡减少,闭锁卵泡增多.Met组和XNTJT-H组小鼠动情周期显著改善,初级卵泡、次级卵泡增加,闭锁卵泡减少.与Control组比较,Model组小鼠血清LH、T、LH/FSH水平升高,FSH水平降低(P<0.01).与Model组比较,Met组与XNTJT-H组小鼠血清LH、T、LH/FSH水平降低,FSH水平升高(P<0.01).与 Control 组比较,Model 组小鼠 p-NF-κB p65/NF-κB p65 比值升高,p-IRS-1/IRS-1、p-PI3K/PI3K、p-AKT/AKT比值降低(P<0.01).与Model组比较,XNTJT-H组小鼠p-NF-κBp65/NF-κB p65 比值降低,p-IRS-1/IRS-1、p-PI3K/PI3K、p-AKT/AKT比值升高(P<0.05).与Control组比较,Model组小鼠血清TNF-α、IL-6 及NF-κB水平升高(P<0.05).与Model组比较,Met组和XNTJT-H组小鼠血清TNF-α、IL-6 及NF-κB水平降低(P<0.05).细胞实验:与Control组比较,Model组细胞p-NF-κBp65/NF-κBp65 比值升高,p-IRS-1/IRS-1、p-PI3K/PI3K、p-AKT/AKT比值降低(P<0.01).与Model组比较,XNTJT组细胞p-NF-κBp65/NF-κBp65 比值降低(P<0.01),p-IRS-1/IRS-1、p-PI3K/PI3K、p-AKT/AKT 比值升高(P<0.05).与 XNTJT 组比较,XNTJT+si-IRS-1 组细胞p-PI3K/PI3K和p-AKT/AKT比值降低,p-NF-κBp65/NF-κBp65 比值升高(P<0.01).与Control组比较,Model组细胞增殖率降低(P<0.01).与Model组比较,XNTJT组细胞增殖率升高(P<0.01).与XNTJT组比较,XNTJT+si-IRS-1 组细胞增殖率降低(P<0.01).Model组细胞凋亡率较Control组升高(P<0.01);XNTJT组细胞凋亡率较Model组降低(P<0.01);XNTJT+si-IRS-1 组细胞凋亡率较XNTJT组升高(P<0.01).Model组细胞TNF-α、IL-6 及NF-κB水平较Control组升高(P<0.01);XNTJT组细胞TNF-α、IL-6 及NF-κB水平较Model组降低(P<0.01);XNTJT+si-IRS-1 组细胞TNF-α、IL-6 及NF-κB水平较XNTJT组升高(P<0.01).结论:消囊调经汤可能通过调控NF-κB/IRS-1/PI3K/AKT通路改善肥胖型PCOS小鼠代谢与生殖异常,调节卵巢颗粒细胞功能.
Objective:To investigate the mechanism of Xiaonang Tiaojing Decoction in treating obese polycystic ovary syndrome(PCOS)by regulating the nuclear factor kappa-B(NF-κB)/insulin receptor substrate-1(IRS-1)/phosphatidylinositol-3-kinase(PI3K)/protein kinase B(AKT)pathway.Methods:An obese PCOS mouse model was established using a high-fat diet combined with dehydroepiandrosterone.Mice were divided into a normal control group(Control),a model group(Model),a metformin group(200 mg·kg-1,Met),a low-dose Xiaonang Tiaojing Decoction group(5 g·kg-1,XNTJT-L),and a high-dose Xiaonang Tiaojing Decoctiion group(20 g·kg-1,XNTJT-H),with10 mice per group,administered orally for8 weeks.Ovarian granulosa cells were di-vided into a blank group(Control),a model group(Model),a drug-containing serum group(XNTJT),a drug-containing serum si-NC group(XNTJT si-NC),and a drug-containing serum si-IRS-1 group(XNTJT si-IRS-1).Mouse insulin resistance in-dex(HOMA-IR)and serum free fatty acids(FFA)levels were measured;HE staining was used to assess mouse estrous cycle and o-varian tissue pathology;ELISA was used to detect serum levels of luteinizing hormone(LH),follicle-stimulating hormone(FSH),tes-tosterone(T),and LH/FSH ratio;Western blot analyzed key protein expression levels of the NF-κB/IRS-1/PI3K/AKT pathway in mice and cells;ELISA measured tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),and NF-κB levels in mice and cells;CCK-8 assay assessed cell proliferation;and flow cytometry analyzed cell apoptosis.Results:Animal experiments:Compared with the Control group,the levels of HOMA-IR and FFA in the Model group were increased(P<0.01).Compared with the Model group,the levels of HOMA-IR and FFA in the Met group and XNTJT-H group were decreased(P<0.05).The estrous cycle of the mice in the Control group was regular,and follicles at different stages of development could be seen.The mice in the model group had an estrotic cy-cle disorder,with a decrease in primary and secondary follicles,and an increase in atresia follicles.The estrous cycle of mice in the Met group and XNTJT-H group was significantly improved,with an increase in primary and secondary follicles,and a decrease in atresia follicles.Compared with the Control group,the serum levels of LH,T,LH/FSH in the Model group were increased,and the FSH level decreased(P<0.01).Compared with the Model group,the serum levels of LH,T,LH/FSH,and FSH in the Met group and XNTJT-H group were decreased,and the FSH level was increased(P<0.01).Compared with the Control group,the p-NF-κB p65/NF-κB p65 ratio of mice in the Model group was increased,and the ratio of p-IRS-1/IRS-1,p-PI3K/PI3K and p-AKT/AKT decreased(P<0.01).Compared with the model group,the ratio of p-NF-κB p65/NF-κB p65 decreased in the XNTJT-H group and in-creased the ratio of p-IRS-1/IRS-1,p-PI3K/PI3K and p-AKT/AKT(P<0.05).Compared with the Control group,the serum levels of TNF-α,IL-6 and NF-κB in the Model group were increased(P<0.05).Compared with the Model group,the serum lev-els of TNF-α,IL-6 and NF-κB in the Met group and XNTJT-H group were decreased(P<0.05).Cell experiments:Compared with the Control group,the p-NF-κB p65/NF-κB p65 ratio of the cells in the Model group was increased,and the p-IRS-1/IRS-1,p-PI3K/PI3K,and p-AKT/AKT ratios were decreased(P<0.01).Compared with the Model group,the p-NF-κB p65/NF-κB p65 ratio of the cells in the XNTJT group decreased(P<0.01),and the ratio of p-IRS-1/IRS-1,p-PI3K/PI3K and p-AKT/AKT increased(P<0.05).Compared with the XNTJT group,the p-PI3K/PI3K and p-AKT/AKT ratios of the cells in the XNTJT si-IRS-1 group decreased,and the p-NF-κB p65/NF-κB p65 ratio increased(P<0.01).Compared with the Control group,the cell proliferation rate of the Model group was reduced(P<0.01).Compared with the Model group,the XNTJT group showed an increased cell proliferation rate(P<0.01).Compared with the XNTJT group,the XNTJT si-IRS-1 group had a decreased cell proliferation rate(P<0.01).The apoptosis rate of cells in the Model group was higher than that in the Control group(P<0.01);the apoptosis rate in the XNTJT group was lower than that in the Model group(P<0.01);the apoptosis rate in the XNTJT si-IRS-1 group was higher than that in the XNTJT group(P<0.01).Levels of TNF-α,IL-6,and NF-κB in cells of the Model group were higher than those in the Control group(P<0.01);levels in the XNTJT group were lower than those in the Model group(P<0.01);levels in the XNTJT si-IRS-1 group were higher than those in the XNTJT group(P<0.01).Conclusion:Xiaonang Tiaojing Decoc-tion may improve metabolic and reproductive abnormalities in obese PCOS mice and modulate ovarian granulosa cell function by regula-ting the NF-κB/IRS-1/PI3K/AKT pathway.
