靶向HMGCS1探讨对宫颈癌细胞线粒体基因表达调控的作用机制OA
Targeting HMGCS1 to explore the mechanism of action on the regulation of mitochondrial gene expression in cervical cancer cells
目的:探讨 3-羟基-3-甲基戊二酰辅酶 A合成酶 1(3-hydroxy-3-methylglutaryl coenzyme A synthase 1,HMGCS1)对宫颈癌(cervical cancer,CC)细胞线粒体基因表达的调控作用机制.方法:利用生物信息学分析、亚细胞结构分离定位法、shRNA慢病毒载体的构建及细胞转染、Western blot、实时荧光定量PCR、RNA合成与降解动力学分析、线粒体DNA免疫沉淀-定量PCR分析,探索靶向HMGCS1调控CC细胞线粒体基因表达的作用机制.结果:HMGCS1在CC中表达上调,预示预后不良;HMGCS1主要差异表达于线粒体中;HMGCS1的敲低导致CC细胞线粒体基因表达水平普遍下调;HMGCS1的敲低可能通过降低线粒体mRNA合成水平来抑制线粒体基因表达;HMGCS1可能通过促进CC细胞线粒体基因转录调控因子与线粒体DNA(mtDNA)的结合来保障线粒体基因的正常转录.结论:HMGCS1在CC中高表达,并与不良预后相关.下调的HMGCS1通过降低CC细胞线粒体mRNA合成水平来抑制线粒体基因表达,并且HMGCS1可通过促进CC细胞线粒体基因转录调控因子与mtDNA的结合来保障线粒体基因的正常转录.因此,靶向抑制HMGCS1调控线粒体基因表达可能是一种很有前景的CC治疗新策略.
Objective:To investigate the mechanism of the regulatory effect of 3-hydroxy-3-methylglutaryl coenzyme A syn-thase 1(HMGCS1)on mitochondrial gene expression in cervical cancer(CC)cells.Methods:Bioinformatics analysis,subcellu-lar structure isolation and localization method,construction of shRNA lentiviral vector and cell transfection,Western blot,real-time fluorescence quantitative PCR,kinetic analysis of RNA synthesis and degradation,and mitochondrial DNA immunoprecipitation-quantitative PCR analysis were utilized to explore the mechanism by which targeting HMGCS1 regulates mi-tochondrial gene expression in CC cells by exploring the mechanism of action of targeting HMGCS1.Results:HMGCS1 expres-sion is up-regulated in CC and predicts poor prognosis;HMGCS1 is mainly differentially expressed in mitochondria;knockdown of HMGCS1 leads to a general down-regulation of the mitochondrial gene expression level in CC cells;knockdown of HMGCS1 may inhibit mitochondrial gene expression by decreasing the level of mitochondrial mRNA synthesis;HMGCS1 may promote the mitochondrial gene transcription of CC cells by regulator binding to mitochondrial DNA(mtDNA)to safeguard the normal tran-scription of mitochondrial genes.Conclusion:HMGCS1 is highly expressed in CC and is associated with poor prognosis.Down-regulated HMGCS1 inhibits mitochondrial gene expression by decreasing the level of mitochondrial mRNA synthesis in CC cells,and HMGCS1 can safeguard the normal transcription of mitochondrial genes by promoting the binding of mitochondrial gene transcriptional regulators to mtDNA in CC cells.Therefore,targeted inhibition of HMGCS1 to regulate mitochondrial gene ex-pression may be a promising new strategy for CC treatment.
