依达拉奉右莰醇对CRS小鼠焦虑抑郁行为的影响和机制研究OA
Research on the impact and mechanism of edaravone dexborneol on anxiety and depression behaviors in CRS mice
目的:探讨依达拉奉右莰醇(edaravone dexborneol,EDB)对慢性约束性应激(chronic restraint stress,CRS)小鼠焦虑抑郁行为的改善作用及其潜在机制.方法:将雄性C57BL/6小鼠随机分配为4组(n=8):对照(Control)组、慢性约束性应激(CRS)组、EDB[CRS+EDB(5 mg/kg)]组,EDB[CRS+EDB(10 mg/kg)]组.通过约束应激小鼠每天4 h,连续21 d,诱导抑郁模型.采用旷场实验、悬尾实验、强迫游泳实验评估小鼠焦虑抑郁行为;qPCR检测GSDMD、IL-1β、AKT、p62、Beclin1、LC3Ⅱ mRNA表达;Western blot检测p-AKT、p-AMPK、p62、Beclin1、LC3B、GSDMD、GSDMD-N、Pro-IL-1β、IL-1β、BDNF蛋白表达;免疫荧光观察海马神经元特异性核蛋白(NeuN)表达并计数.结果:旷场实验中CRS组小鼠运动总距离、中心区停留时间、平均运动速度均明显低于Control组(P<0.05),EDB给药各组运动总距离、中心区停留时间、平均运动速度均高于CRS组(P<0.05);悬尾试验和强迫游泳实验中CRS组小鼠不动时间明显高于Control组(P<0.05),EDB各组不动时间明显低于CRS组(P<0.05);qPCR结果显示,与Control组相比,CRS组GSDMD、IL-1β、p62 mRNA表达增加,Beclin1、LC3Ⅱ mRNA表达减少(P<0.05);与CRS组相比,EDB各组GSDMD、IL-1β、p62 mRNA表达减少,Beclin1、LC3Ⅱ mRNA表达增加(P<0.05);Western blot结果显示,与Control组相比,CRS组p-AKT、p62、GSDMD、GSDMD-N、Pro-IL-1β、IL-1β表达增加,p-AMPK、Beclin1、LC3B、BDNF表达减少(P<0.05);与CRS组相比,EDB各组p-AKT、p62、GSDMD、GSDMD-N、Pro-IL-1β、IL-1β表达减少,p-AMPK、Beclin1、LC3B、BDNF表达增加(P<0.05);免疫荧光结果显示,海马CA1、CA3、DG区CRS组NeuN数量较Control组明显减少(P<0.05),EDB各组NeuN数量较CRS组明显增加(P<0.05).结论:EDB可能通过AKT/AMPK通路调节细胞自噬,抑制焦亡及炎症反应,减轻神经元的损伤,改善CRS诱导的小鼠焦虑抑郁行为.
Objective:To investigate the effects of edaravone borneol(EDB)on anxiety and depressive behaviors in chronic restraint stress(CRS)mice and its potential mechanisms.Methods:Male C57BL/6 mice were randomly assigned to 4 groups(n=8):Control group,Chronic restraint stress(CRS)group,EDB[CRS+EDB(5 mg/kg)]group,and EDB[CRS+EDB(10 mg/kg)]group.The depression model was induced by restraining the mice for 4 hours daily for 21 consecutive days.Anxiety and depressive behaviors were assessed using the open field test,tail suspension test,and forced swimming test.qPCR was used to de-tect the expression of GSDMD,IL-1β,AKT,p62,Beclin1,and LC3Ⅱ mRNA.Western blot was employed to measure the pro-tein expression of p-AKT,p-AMPK,p62,Beclin1,LC3B,GSDMD,GSDMD-N,Pro-IL-1β,IL-1β,and BDNF.Immunoflu-orescence was used to observe and count the expression of the neuronal-specific nuclear protein(NeuN)in the hippocampus.Re-sults:In the open field test,the total distance travelled,time spent in the center zone,and average speed of movement in CRS mice were significantly lower than those in the Control group(P<0.05),while these parameters were higher in EDB-treated groups compared to the CRS group(P<0.05).In the tail suspension and forced swimming tests,the immobility time in CRS mice was significantly longer than that in the Control group(P<0.05),while it was shorter in EDB-treated groups compared to the CRS group(P<0.05).qPCR results showed that compared to the Control group,the expression of GSDMD,IL-1β,and p62 mRNA increased,while the expression of Beclin1 and LC3Ⅱ mRNA decreased in the CRS group(P<0.05).Compared to the CRS group,the expression of GSDMD,IL-1β,and p62 mRNA decreased,while the expression of Beclin1 and LC3Ⅱ mRNA in-creased in EDB-treated groups(P<0.05).Western blot results indicated that compared to the Control group,the expression of p-AKT,p62,GSDMD,GSDMD-N,Pro-IL-1β,and IL-1β increased,while the expression of p-AMPK,Beclin1,LC3B,and BDNF decreased in the CRS group(P<0.05).Compared to the CRS group,the expression of p-AKT,p62,GSDMD,GSDMD-N,Pro-IL-1β,and IL-1β decreased,while the expression of p-AMPK,Beclin1,LC3B,and BDNF increased in EDB-treated groups(P<0.05).Immunofluorescence results showed that the number of NeuN in the CA1,CA3,and DG regions of the hippocampus was significantly lower in the CRS group than in the Control group(P<0.05),while it was significantly high-er in EDB-treated groups compared to the CRS group(P<0.05).Conclusion:EDB may improve anxiety and depressive behav-iors in CRS-induced mice by regulating autophagy through the AKT/AMPK pathway,inhibiting pyroptosis and inflammatory re-sponses,and reducing neuronal damage.
