以肺形侧耳菌株为试材,采用菌落形态与菌丝显微观察、对峙培养法、ITS序列分析及平板培养等方法,研究了不同阔叶树凋落物作为栽培基质对肺形侧耳菌丝生长、子实体产量及品质的影响,以期为拓展肺形侧耳低成本栽培基质的选择范围及凋落物资源化利用提供参考依据.结果表明:在7种阔叶树凋落物基质上,DL-P.pul001菌株展现出明显的菌丝生长速度优势,且在紫椴凋落物上生长速度最快((4.02±0.19)mm·d-1),显著快于木屑对照((3.10±0.05)mm·d-1);进一步以紫椴凋落物为主要基质,设计了 4种配方栽培肺形侧耳DL-P.pul001,配方Ⅲ(55%凋落物、18%菌糠、15%麦麸、10%玉米芯、1%石灰、1%石膏)栽培出的肺形侧耳在子实体产量((133.00±1.55)g·袋-1)、菌柄直径((2.24±0.27)cm)、菌盖长度((7.21±0.54)cm)、菌盖宽度((6.57±0.36)cm)及粗蛋白((235.75±0.27)mg·g-1)均优于木屑对照;在粗多糖((19.15±0.18)mg·g-1)和粗脂肪(2.98%±0.09%)产出配方Ⅱ(65%凋落物、8%菌糠、15%麦麸、10%玉米芯、1%石灰、1%石膏)表现最好.

The effects of different broad-leaved tree litter on mycelial growth,fruiting body yield and quality of Pleurotus pulmonata were studied by microscopic observation of colony morphology and hyphae,stand-off culture,ITS sequence analysis and plate culture with P.pulmonata strains as test materials,in order to provide reference for expanding the selection range of low-cost cultural substrates and resource utilization of litter of P.pulmonata.The results showed that DL-P.pul001 showed obvious superiority in hyphae growth rate on 7 broad-leaved tree litter substrates,and the growth rate on Tilia amurensis litter was the fastest((4.02±0.19)mm·d-1),significantly faster than that of sawdust control((3.10±0.05)mm·d-1).Furthermore,four formulas were designed to cultivate P.pulmonata DL-P.pul001 with T.amurensis litter as the main substrate.The fruit body yield((133.00±1.55)g·bag-1),stipe diameter((2.24±0.27)cm),capsule length((7.21±0.54)cm),capsule width((6.57±0.36)cm)and crude protein((235.75±0.27)mg·g-1)of P.pulmonata cultivated with formula Ⅲ(55%litter,18%fungus bran,15%wheat bran,10%corncob,1%lime,1%gypsum)were better than those of the sawdust control group.Formula Ⅱ(65%litter,8%fungus chaff,15%wheat bran,10%corncob,1%lime,1%gypsum)was the best in the yield of crude polysaccharide((19.15±0.18)mg·g-1)and crude fat(2.98%±0.09%).
