以栽培种辣椒(Capsicum annuum L.)为试材,采用生物信息学分析的方法,研究了辣椒基因组水平上的KNOX基因,KNOX基因家族成员的理化性质、基因结构、蛋白质保守结构域、系统进化、启动子顺式作用元件以及组织与逆境表达模式,以期为KNOX基因在辣椒生长发育、逆境胁迫响应中的功能提供参考依据.结果表明:辣椒基因组中共有7个KNOX基因家族成员,分布在7条染色体上,绝大多数编码酸性蛋白,均为亲水性蛋白,并全部定位于细胞核.系统发育分析可将辣椒KNOX基因家族分为Class Ⅰ和Class Ⅱ2个亚家族,并进一步分为5个亚组.启动子分析发现一些与生长发育、激素和胁迫响应相关的顺式作用调控元件.组织表达分析结果显示,Class Ⅰ类基因表达组织特异性明显,主要在根、茎、叶等组织中高表达;Class Ⅱ类基因时空表达更广泛.在7种非生物胁迫处理下,部分基因表达量变化较大,其中5个基因响应特定胁迫.RT-qPCR验证了 2个特异性响应冷胁迫、盐胁迫、热胁迫的基因.

Taking cultivated pepper(Capsicum annuum L.)as the test material,bioinformatics analysis was used to study the KNOX gene at the pepper genome level,including the physicochemical properties,gene structure,protein conserved domains,systematic evolution,promoter cis acting elements,and tissue and stress expression patterns of KNOX gene family members,in order to provide reference for the function of KNOX gene in pepper growth and development,as well as stress response.The results showed that there were a total of 7 KNOX gene family members in the pepper genome,distributed on 7 chromosomes.The majority encoded acidic proteins,were all hydrophilic proteins,and were all located in the nucleus.Phylogenetic analysis could divide the pepper KNOX family into two subfamilies,Class Ⅰ and Class Ⅱ,and further into five subgroups.Promoter analysis revealed some cis-acting regulatory elements related to growth and development,hormones,and stress responses.Tissue expression analysis showed that the Class Ⅰ genes had obvious tissue specificity and were mainly highly expressed in roots,stems,and leaves;the Class Ⅱ genes had a more extensive spatiotemporal expression.Under seven abiotic stress treatments,the expression levels of some genes changed significantly,among which five genes responded to specific stresses.RT-qPCR verified two genes that specifically responded to cold stress,salt stress,and heat stress.
