五点采样法评估安格斯牛眼肌脂肪酸与蛋白表达优势研究OA
An assessment of fatty acid and protein expression dominance in longissimus dorsi muscle of angus cattle via a five-point sampling method
该研究旨在比较单点采样和五点采样方法对安格斯肉牛脂肪酸组成、肌内脂肪沉积相关蛋白表达的影响,为国内牛肉品质评估提供科学依据,并为肉牛育种和肉质改良提供参考.该试验分为 2 个部分,第一部分先随机选取3头 26月龄的安格斯阉牛,屠宰后每头牛于眼肌部位分别使用单点和五点采样方法取样进行脂肪酸测定,并对SCD、SREBP1、PPA R γ、C/EBPΩ、FA B P4、ACACA 以及FA SN进行Western blot检测,第二部分选取 30 头 26月龄的安格斯阉牛,以单点采样和五点采样方法随机分为2组各15 头,屠宰后分析安格斯肉牛大理石花纹和屠宰性能的影响差异,后于眼肌处取样再次进行脂肪酸测定.结果表明,试验一的单点采样组中单不饱和脂肪酸的肉豆蔻烯酸、棕榈油酸,多不饱和脂肪酸的亚油酸以及γ-亚麻酸含量显著低于五点采样组(P<0.05),而饱和脂肪酸中辛酸的含量显著高于五点采样组(P<0.05).在蛋白表达水平上,五点采样组的PPARγ、C/EBPΩ以及SCD表达量显著高于单点采样组(P<0.05),而在SREBP1、FA BP4、ACACA 以及FA SN表达上无显著差异(P>0.05),但具有生物学上升趋势.试验二的单点采样组饱和脂肪酸以及n-6/n-3脂肪酸显著高于五点采样组(P<0.05),而不饱和脂肪酸、单不饱和脂肪酸/饱和脂肪酸、多不饱和脂肪酸/饱和脂肪酸、n-6脂肪酸、n-3脂肪酸显著低于五点采样组(P<0.05).因此,与单点采样法相比,五点采样法能更全面地反映样本的脂肪酸组成及相关蛋白表达特征,这一优势主要与其可检测到更高的不饱和脂肪酸含量、更优的n-6/n-3 脂肪酸比例,以及PPA R γ、C/EBPΩ和SCD蛋白表达水平上调的结果密切相关.
The aim of this study was to compare the effects of single-point and five-point sampling methods on the fatty acid compo-sition and expression of intramuscular fat deposition-related proteins in Angus beef cattle,to provide a scientific basis for the evalu-ation of beef quality in China,and to provide a reference for the breeding of beef cattle and the improvement of meat quality.The experiment was divided into two parts.In the first part,three 26-month-old Angus steers were randomly selected,and after slaugh-ter,each cow was sampled in the eye muscle area for fatty acid determination using single-point and five-point sampling methods,respectively,and Western blot for SCD,SREBP1,PPARγ,C/EBPΩ,FABP4,ACACA and FA SN,and in the second part,thirty 26-month-old Angus steers were randomly selected.Thirty 26-month-old Angus beef cattle were randomly divided into 2 groups of 15 cattle each by single-point sampling and five-point sampling methods,and the differences in the effects of marbling and slaugh-ter performance of Angus beef cattle were analyzed after slaughter,and samples were taken from the eye muscles for fatty acid as-say again afterward.In the single-point sampling group of Experiment 1,the levels of monounsaturated fatty acids(macadamia acid and palmitic acid)and polyunsaturated fatty acids(linoleic acid and γ-linolenic acid)were significantly lower than those in the five-point sampling group(P<0.05).Conversely,the level of saturated fatty acid(caprylic acid)was significantly higher than that in the five-point sampling group(P<0.05).At the protein expression level,the five-point sampling group exhibited significantly higher expression levels of PPARγ,C/EBPΩ,and SCD compared to the single-point sampling group(P<0.05).No significant differences were observed in the expression of SREBP1,FA BP4,A CA CA,and FASN(P﹥0.05),though a biological upward trend was noted.In Experiment 2,the single-point sampling group exhibited significantly higher saturated fatty acids and n-6/n-3 fatty acid ratios than the five-point sampling group(P<0.05).However,it showed significantly lower levels of unsaturated fatty acids,monounsaturated fatty acids/saturated fatty acids ratio,polyunsaturated fatty acids/saturated fatty acids ratio,n-6 fatty acids,and n-3 fatty acids com-pared to the five-point sampling group(P<0.05).Therefore,compared to single-point sampling,five-point sampling provides a more comprehensive reflection of the fatty acid composition and related protein expression characteristics in the sample.This advantage is primarily associated with its ability to detect higher unsaturated fatty acid content,a more favorable n-6/n-3 fatty acid ratio,and the upregulation of PPA Rγ,C/EBPΩ,and SCDprotein expression levels.
