为明确肌球蛋白重链(MyH)家族基因的特征与功能,该研究分析了其多个亚型在信阳水牛不同部位肌肉组织中的表达差异.通过对MyH家族基因进行motif和结构域分析,构建系统进化树摸清进化关系,PPI网络互作图、GO富集分析和KEGG富集分析掌握基因家族功能,检测其在各部分肌肉组织中的表达情况,了解信阳水牛的肌肉发育特征.结果显示,MyH 基因家族含有 3 个motif和 3 个保守结构域,进化上MyH1、MyH2 以及MyH4亲缘关系较近,富集分析发现MyH基因家族主要富集在肌动蛋白细胞骨架调节、紧密连接及血管平滑肌收缩等信号通路.组织表达谱结果表明MyH1、MyH2 和MyH4 在前腿肌和后腿肌中的表达量较高,MyH7 在背最长肌中的表达量最高.综上,该研究揭示了信阳水牛MyH基因家族的分子特征、进化关系及功能富集通路,并明确了其在不同肌肉部位的表达模式.

In order to further clarify the myosin heavy chain(MyH)family gene characteristics and gene functions,and to analyze the expression of its multiple isoforms in various parts of muscle tissues of Xinyang buffalo.In this study,we analyzed the motif and structural domains of the MyH family genes and mapped the evolutionary relationships by constructing a phylogenetic tree,grasped the functions of the gene family through the PPI network interactions map,GO enrichment analysis and KEGG enrichment analysis,and understood the muscle developmental characteristics of Xinyang buffaloes by detecting their expression in various parts of mus-cle tissues.The results showed that the MyH gene family contains three motifs and three conserved structural domains,and evolu-tionarily MyH1,MyH2 and MyH4 are relatively close to each other.Enrichment analysis revealed that the MyH gene family is main-ly enriched in the signaling pathways of actin cytoskeleton regulation,tight junctions and vascular smooth muscle contraction.Tis-sue expression profiling showed that MyH1,MyH2 and MyH4 were highly expressed in the foreleg and hind leg muscles,and MyH7 was the most highly expressed in the longissimus dorsi muscle.In summary,this study revealed the molecular characteristics,evolu-tionary relationships and functional enrichment pathways of the MyH gene family in Xinyang buffalo,and clarified its expression patterns in different muscle parts.
