基于NF-κB/I-κB通路探讨坤灵丸改善DOR大鼠卵巢功能机制OA
Mechanism of Kunling Pill in Improving Ovarian Function in DOR Rats Via the NF-κB/I-κB Pathway
目的 探讨坤灵丸通过调节核因子-κB(nuclear factor-κB,NF-κB)/核因子-κB抑制蛋白(inhibitor of nuclear factor-κB,I-κB)信号通路,改善环磷酰胺(cyclophosphamide,CTX)诱导的卵巢储备功能下降(diminished ovarian reserve,DOR)大鼠卵巢炎症损伤的作用机制.方法 将45只具有正常动情周期的SD雌鼠随机分为正常对照组、模型组、阳性药治疗组(戊酸雌二醇)、坤灵丸低剂量组(0.5 g/kg)和坤灵丸高剂量组(1 g/kg),每组9只.除正常对照组外,其余各组大鼠均单次腹腔注射CTX(90 mg/kg)以建立大鼠DOR模型.连续灌胃给药28 d后,检测血清激素雌二醇(estradiol,E2)、抗苗勒管激素(anti-Müllerian hormone,AMH)、卵泡刺激素(follicle-stimulating hormone,FSH)、炎症因子白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-4(interleukin-4,IL-4)、白细胞介素-6(interleukin-6,IL-6)水平,并检测卵巢组织NF-κB/I-κB蛋白表达.结果 与模型组比较,坤灵丸低剂量组和坤灵丸高剂量组血清AMH、E2均明显升高,FSH水平无明显改变.坤灵丸能降低DOR大鼠血清IL-1β、IL-6水平,并下调卵巢组织中NF-κB、I-κB蛋白的表达.结论 坤灵丸可以改善CTX诱导的DOR大鼠卵巢储备功能.其作用机制可能通过抑制NF-κB/I-κB信号通路介导炎症反应,减轻炎症对卵巢的损伤,从而改善大鼠卵巢功能.
Objective To investigate the mechanism by which Kunling Pill improves cyclophosphamide(CTX)-induced diminished ovarian reserve(DOR)in rats by regulating the nuclear factor kappa B(NF-κB)/inhibitor of nuclear factor kappa B(I-κB)signaling pathway.Methods 45 Sprague Dawley(SD)female rats with normal estrous cycles were randomly divided into a normal control group,a model group,a positive drug group(estradiol valerate),a low-dose Kunling Pill group(0.5 g/kg),and a high-dose Kunling Pill group(1 g/kg),with 9 rats in each group.Except for the normal control group,all groups were established with a single injection of CTX(90 mg/kg)to establish a rat DOR model.After 28 days of continuous oral administration,serum hormones estradiol(E2),anti-Müllerian hormone(AMH),follicle stimulating hormone(FSH),inflammatory factors interleukin-1 beta(IL-1 β),interleukin-4(IL-4),interleukin-6(IL-6),and ovarian tissue NF-κB/IκB protein expression were detected.Results As a result,Kunling Pill can regulate the hormone levels in rats.Compared with the model group,the low-dose Kunling Pill group and high-dose Kunling Pill group showed significant increases in serum AMH and E2,while FSH showed no significant changes.Kunling Pill reduces the accumulation of IL-1β and IL-6 in the serum of DOR rats,and lowers the levels of NF-κB and I-κB to alleviate inflammatory reactions.Conclusion Kunling Pill can improve ovarian reserve function.The mechanism may be mediated by inhibiting the NF-κB/I-κ B pathway to alleviate inflammation induced ovarian damage and improve rat ovarian function.
