基于JAK2/STAT3通路探讨丹红注射液对慢性心力衰竭大鼠心肌细胞凋亡的影响OA
Exploration on the Effects of Danhong Injection on Myocardial Cell Apoptosis in Rats with Chronic Heart Failure Based on JAK2/STAT3 Pathway
目的 基于JAK2/STAT3通路探讨丹红注射液对慢性心力衰竭(CHF)大鼠心肌细胞凋亡的影响及作用机制.方法 采用腹主动脉缩窄术建立慢性心力衰竭大鼠模型,采用随机数字表法将成模大鼠分为模型组、丹红注射液组和卡托普利组,另设假手术组,各组大鼠分别予相应药液灌胃,连续15 d.心脏彩超检测大鼠心室结构及心功能指标[左室舒张末内径(LVEDD)、左室收缩末内径(LVESD)、左室射血分数(LVEF)、左室短轴缩短率(LVFS)],ELISA检测血清氨基末端脑钠肽前体(NT-proBNP)、肿瘤坏死因子(TNF)-α和白细胞介素(IL)-6含量,HE和Masson染色观察心肌组织形态和心肌纤维化情况,TUNEL法检测心肌细胞凋亡率,RT-qPCR检测心肌组织Janus酪氨酸激酶2(JAK2)、信号转导和转录激活因子3(STAT3)、Bcl-2、Bax、Caspase-3、Caspase-9 mRNA表达,Western blot检测p-JAK2、JAK2、p-STAT3、STAT3、Bcl-2、Bax、Caspase-3、Caspase-9蛋白表达.结果 与假手术组比较,模型组大鼠萎靡不振,活动迟缓,毛发暗沉干燥易脱落,爪甲、舌质紫黯;心脏彩超显示LVEDD、LVESD显著增加(P<0.01),LVEF、LVFS显著降低(P<0.01);血清NT-proBNP、TNF-α、IL-6含量显著升高(P<0.01);心肌细胞排列紊乱,伴细胞肥大、坏死,胶原纤维增生,炎性细胞浸润及心肌纤维化率升高(P<0.01),心肌细胞凋亡率显著升高(P<0.01),心肌组织JAK2、STAT3 mRNA和p-JAK2、p-STAT3 蛋白表达显著升高,Bax、Caspase-3、Caspase-9 mRNA和蛋白表达显著升高,Bcl-2 mRNA和蛋白表达显著降低(P<0.01).与模型组比较,丹红注射液组和卡托普利组大鼠精神状态、饮食及活动能力改善,毛发有光泽,爪甲、舌质颜色恢复;LVEDD、LVESD显著减少,LVEF、LVFS显著升高(P<0.01);血清NT-proBNP、TNF-α、IL-6含量显著降低(P<0.01);心肌组织结构轻度异常,胶原纤维增生和心肌纤维脂肪变性减少,心肌纤维化率显著降低(P<0.01),心肌细胞凋亡率显著下降(P<0.01);心肌组织JAK2、STAT3 mRNA和p-JAK2、p-STAT3蛋白表达显著降低,Bax、Caspase-3、Caspase-9 mRNA和蛋白表达显著降低,Bcl-2 mRNA和蛋白表达显著升高(P<0.01).结论 丹红注射液可能通过抑制JAK2/STAT3通路减轻心肌细胞凋亡,改善CHF模型大鼠心肌组织结构损伤,促进心功能恢复.
Objective To explore the effects and mechanism of Danhong Injection in cardiomyocyte apoptosis in rats with chronic heart failure(CHF)based on JAK2/STAT3 pathway.Methods A rat model of CHF was established by abdominal aortic narrowing.The modeled rats were divided into the model group,Danhong Injection group and captopril group according to random number table method,the sham-operation group was set up.The rats in each group were given corresponding drug by gavage for 15 consecutive days.Cardiac ultrasound was used to detect the ventricular structure and cardiac function indexes(LVEDD,LVESD,LVEF,LVFS),serum NT-proBNP,TNF-α and IL-6 were detected by ELISA,HE and Masson staining were used to observe the pathological changes of myocardial tissue and the characteristics of myocardial fibers,apoptosis rate of cardiomyocytes was detected by TUNEL method,RT-qPCR was used to detect the expressions of JAK2,STAT3,Bcl-2,Bax,Caspase-3 and Caspase-9 mRNA in myocardial tissue,the protein expressions of p-JAK2,JAK2,p-STAT3,STAT3,Bcl-2,Bax,Caspase-3 and Caspase-9 was detected by Western blot.Results Compared with the sham-operation group,rats in the model group were depressed,slow in activity,and had dark,dry and easy-to-lose hair,and purple paw nails and tongue;cardiac ultrasound showed a significant increase in LVEDD and LVESD(P<0.01),and a significant decrease in LVEF and LVFS(P<0.01);the contents of serum NT-proBNP,TNF-α and IL-6 significantly increased(P<0.01);cardiomyocytes arrangement disturbed with cellular hypertrophy,necrosis,collagen fiber hyperplasia,inflammatory cell infiltration and increased myocardial fibrosis(P<0.01),the apoptosis rate of cardiomyocytes was increased(P<0.01),the expressions of JAK2,STAT3 mRNA and p-JAK2,p-STAT3 protein in myocardial tissue significantly increased,the expressions of Bax,Caspase-3,Caspase-9 mRNA and protein significantly increased,and the expressions of Bcl-2 mRNA and protein significantly decreased(P<0.01).Compared with the model group,rats in Danhong Injection group and captopril group showed improved mental status,diet and activity,shiny hair,and red paw nails and tongue;LVEDD and LVESD significantly reduced,and LVEF and LVFS increased(P<0.01);the contents of NT-proBNP,TNF-α and IL-6 significantly decreased(P<0.01);myocardial tissue structure was slightly abnormal,collagen fiber proliferation and myocardial fiber steatosis were reduced,myocardial fibrosis rate was significantly decreased(P<0.01),the apoptosis rate of cardiomyocytes significantly decreased(P<0.01);the expressions of JAK2,STAT3 mRNA and p-JAK2,p-STAT3 protein in myocardial tissue significantly decreased,the expressions of Bax,Caspase-3,Caspase-9 mRNA and protein significantly decreased,and the expressions of Bcl-2 mRNA and protein significantly increased(P<0.01).Conclusion Danhong Injection may reduce cardiomyocyte apoptosis by inhibiting JAK2/STAT3 pathway,improve myocardial tissue damage and promote the recovery of cardiac function in CHF rats.
