旨在探究猫传染性腹膜炎病毒(FIPV)刺激对猫外周血单个核细胞(PBMC)免疫功能及磷酸二酯酶(PDE)相关基因表达的影响,为揭示FIPV引起的相关免疫机制和临床中药治疗积累数据.首先,分离猫PBMC并分为对照组、低浓度FIPV组和高浓度FIPV组.通过流式细胞术、酶联免疫吸附试验(ELISA)观察FIPV刺激下免疫细胞、免疫球蛋白和一些免疫因子的变化;利用实时荧光定量PCR检测FIPV刺激下不同质量浓度虫草素与甘露消毒丹(GXM)对CRFK细胞和PBMC内的磷酸二酯酶1B(PDE1B)基因、磷酸二酯酶4B(PDE4B)基因相对表达量的影响;通过ELISA检测FIPV刺激下不同质量浓度虫草素与GXM作用后CRFK细胞和PBMC内环磷酸腺苷(cAMP)的含量.结果显示,低浓度的FIPV病毒液对CD4+T细胞表现出一定的抑制作用,高剂量时转为促进作用.随着病毒量的递增,CD8+T细胞受到显著抑制且CD4+/CD8+比值显著增高;FIPV刺激下PBMC表现出显著的免疫球蛋白M水平上调,免疫球蛋白G水平下调;FIPV刺激后,PBMC分泌肿瘤坏死因子-α、白细胞介素-4增多,白细胞介素-2、白细胞介素-6减少;FIPV能通过下调PDE4B、PDE1 B基因的表达来降低cAMP水平,而GXM和虫草素能上调两者的表达水平,促进cAMP恢复正常水平.综上所述,低浓度的FIPV刺激下细胞免疫较强于体液免疫,高浓度FIPV则相反;FIPV能降低PDE4B、PDE1B基因表达水平并减少cAMP的分泌;GXM和虫草素能上调PDE4B、PDE1B基因表达水平并促进cAMP恢复正常水平,有望成为临床猫传染性腹膜炎治疗药物.

This study aimed to investigate the effects of feline infectious peritonitis virus(FIPV)stimulation on immunity and phosphodiesterase(PDE)related gene expression of peripheral blood mononuclear cells(PBMCs)in cats,accumulating data for the related immune responses caused by FIPV and the clinical application of traditional Chinese medicine.Firstly,feline PBMCs were isolated and extracted,and then divided into three groups:the control group,the low-concentration FIPV group,and the high-concentration FIPV group.The effects of FIPV stimulation on cat kidney cells(CRFK)and PBMCs were observed through flow cytometry and enzyme-linked immunosorbent assay(ELISA).The changes in immune cells,immunoglobulins,and some immune factors under viral stimulation were observed using real-time fluorescence quantitative PCR to detect the effects of different concentrations of cordycepin and Ganlu Xiaodu Micropill(GXM)on the relative expression levels of PDE1B and PDE4B in CRFK cells and PBMCs under FIPV stimulation.ELISA was used to detect the changes in cAMP levels in CRFK cells and PBMCs stimulated by FIPV at different concentrations of cordycepin and GXM.The results showed that low concentrations of FIPV solution exhibited a certain inhibitory effect on CD4+T cells,which turned into a promoting effect at high doses.As the viral load increases,CD8+T cells were significantly inhibited and the CD4+/CD8+ratio increases significantly.Under FIPV stimulation,PBMCs significantly upregulated the IgM levels and downregulated the IgG levels.After stimulation with FIPV,PBMC secretion of TNF-α and IL-4 increased,while IL-2 and IL-6 decreased.Mechanistically,FIPV reduced cAMP levels by downregulating the gene expression of PDE4B and PDE1B,while GXM and cordycepin upregulating the expression of PDE4B and PDE1B and promoting the restoration of cAMP to normal levels.In summary,under the low dose of FIPV stimulation,cellular immunity is stronger than humoral immunity,while at high concentrations,the opposite is true.FIPV can reduce the expressions of PDE4B and PDE1B genes and decrease the secretion of cAMP.GXM and cordycepin can upregulate the expressions of PDE4B and PDE1B genes and promote the return of cAMP to normal levels,which is expected to become clinical FIP treatment drugs.
