试验旨在优化超临界CO2萃取葡萄皮渣中白藜芦醇提取工艺并研究其抗氧化活性.以白藜芦醇提取率为考察指标,以萃取压力、萃取时间、萃取温度、夹带剂流速为考察单因素,采用Box-Behnken设计试验,采用响应面分析法对提取工艺条件进行优化,同时通过提取物对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基清除率、2,2'-连氮基-双-(3-乙基苯并噻唑啉-6-磺酸)[2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate),ABTS]阳离子自由基清除率分析其抗氧化活性.结果显示:最佳提取工艺为萃取压力25 MPa、萃取时间1.7 h、萃取温度45℃、夹带剂流速0.7 mL/min,此条件下白藜芦醇得率为9.09%,与模型预测值相符;抗氧化试验结果显示,葡萄皮渣提取物具有较强的抗氧化能力,白藜芦醇对DPPH自由基、ABTS自由基的清除率随其浓度的升高而提高,半数抑制浓度(IC50)分别为0.52、0.40 mg/mL.因此,超临界CO2萃取葡萄皮渣中白藜芦醇工艺稳定可行,提取物作为饲料添加剂用于动物养殖很有潜力.

The experiment was to study optimal extraction process andantioxidant activity of resveratrol from grape peel residue by supercritical CO2.The extraction rate of resveratrol was used as the investigation index,extraction pressure,extraction time,extraction temperature and entrainer flow rate were used as the single factors,Box-Behnken design and response surface were used to optimize the extraction process condi-tions.The antioxidant activity of extract was analyzed in terms of 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical clearance and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonate(ABTS)radical cation clearance.Results showed that the optimum extraction conditions were as follows:extraction pressure of 25 MPa,extrac-tion time of 1.7 h,extraction temperature of 45℃and entrainer flow rate of 0.7 mL/min.Under these conditions,the yield of resveratrol was 9.09%.The antioxidant test showed that the extract from grape peel residue had strong antioxidant capacity.The scavenging rates of resveratrol on DPPH and ABTS free radicals was concentration-dependent,and the IC50 was 0.52 mg/mL and 0.40 mg/mL,respec-tively.Therefore,extraction of resveratrol from grape peel by supercritical CO2 is stable and fea-sible,and the extractive is very potential as feed additives for animals breeding.
