化学农药对烤烟根际土壤细菌群落结构与功能的影响OA
Effects of Chemical Pesticides on Structure of Bacterial Community and Function in Rhizosphere Soil of Flue-cured Tobacco
为探究化学农药对根际土壤细菌群落结构及其功能的影响,以烤烟品种KRK26成熟期的根际土壤微生物为研究对象,利用高通量测序技术,分析使用精甲霜·锰锌后烤烟根际土壤细菌群落结构与功能变化.结果表明,使用化学农药(CF)的根际土壤细菌群落OTU数目在各分类单位下均低于对照(CK);Chao指数、Ace指数、Shannon指数、Simpson指数均以CK显著较高;群落组成在门水平上以变形菌门、放线菌门为主,CK、CF合计占比分别为61.84%、71.44%,厚壁菌门、芽单胞菌门、黏球菌门在不同处理之间差异显著,厚壁菌门以CF相对丰度较高,芽单胞菌门、黏菌门均以CK相对丰度较高;在属水平上以鞘氨醇单胞菌属(Sphingomonas)、链霉菌属(Streptomyces)丰度较高,在CF处理中检测出不动杆菌属(Acinetobacter),而CK中没有,拟无枝酸菌属(Amycolatopsis)、芽单胞菌属(Gemmatimonos)在不同处理之间差异显著,且均以CK相对丰度较高;CK、CF根际细菌群落模块化指数分别为0.666 6、0.298 9,同时CK处理节点数和聚集系数均高于CF处理.细菌群落功能共涉及代谢和遗传信息处理等6类一级功能,代谢为最主要功能,CK、CF代谢功能相对丰度分别为70.52%、70.23%;涉及42类二级功能,以碳水化合物代谢、氨基酸代谢为主,CK、CF碳水化合物代谢功能丰度分别为15.88%、15.64%,氨基酸代谢功能丰度分别为14.92%、14.89%,均以CK较高.可见,使用化学农药降低了土壤微生物多样性及稳定性,改变了群落组成,降低了代谢功能丰度.
To investigate the effects of chemical pesticides on the structure and function of bacterial communities in rhizosphere soil,the mature microorganisms in rhizosphere soil of tobacco variety KRK26 were studied.High throughput sequencing was used to analyze the changes in composition,structure,and function of soil bacteria after the application of Metalaxyl-M mancozeb WG.The results showed that the number of OTUs in the bacterial community of rhizosphere soils using chemical pesticides(CF)was lower than that of the control(CK)in all taxonomic units.The Chao index,Ace index,Shannon index,and Simpson index were significantly higher compared to the CK.At the phylum level,the community composition was mainly dominated by Proteobacteria and Actinobacteria,with CK and CF ac-counting for 61.84%and 71.44%respectively.The phyla Firmicutes,Gemmatimonadota,and Myxomycota show significant differences between different treatments,with CF being the most abundant in Firmicutes and CK being the most abundant in Bacteromonas and Myx-omonas.At the genus level,Sphingomonas and Streptomyces have higher abundance.Acinetobacter was found in CF treatment but not in CK.The genera Amycolatopsis and Gemmatimonas showed significant differences between different treatments,with CK being relatively more abundant.The modularity indices for bacterial communities of rhizosphere in CK and CF were 0.666 6 and 0.298 9,respectively.At the same time,the number of nodes and aggregation coefficient in CK treatment were higher than those in CF treatment.The bacterial community function involved in six primary functions such as metabolism and genetic information processing.Metabolism was the most im-portant function and the relative abundance of metabolic functions by CK and CF was 70.52%and 70.23%,respectively.Meanwhile,the bacterial community function involved in 42 secondary functions,mainly carbohydrate metabolism and amino acid metabolism.The a-bundance of carbohydrate metabolic functions by CK and CF was 15.88%and 15.64%,respectively,and the abundance of metabolic functions by amino acid was 14.92%and 14.89%,respectively,all of which were higher in CK.It could be seen that the use of chemi-cal pesticides reduced diversity and stability of microbial in soil,alters community composition,and decreased metabolic functional abun-dance.
