理冲生髓饮治疗卵巢癌作用及机制研究OA

目的:探讨理冲生髓饮通过调控三方基序蛋白44(TRIM44)介导核因子-κB(NF-κB)通路发挥抗卵巢癌(OC)作用的相关分子机制.方法:构建TRIM44稳定过表达人卵巢癌细胞(SKOV3),将细胞接种于Balb/C裸鼠皮下制备移植瘤模型,将40只裸鼠随机分别为空白组、顺铂组、中药组、联合组,每组10只.中药组予理冲生髓饮7.52 g/kg灌胃,2次/日;顺铂组予0.1 mg/mL顺铂溶液腹腔注射,隔日1次;联合组予理冲生髓饮7.52g/kg灌胃,2次/日,同时联合0.1 mg/mL顺铂溶液腹腔注射,隔日1次;空白组予等体积生理盐水灌胃,2次/日.给药干预14d,观察裸鼠的肿瘤瘤体体积及生长趋势;苏木精-伊红(HE)染色观察瘤体组织病理形态学变化;逆转录聚合酶链反应(RT-PCR)检测瘤体中抑制因子κB激酶β(IKKβ)、核因子-κB的抑制蛋白α(IκBα)、核因子-κB p65(NF-κB p65)mRNA表达情况;免疫荧光双染检测瘤体中TRIM44、NF-κB p65蛋白分布情况;蛋白免疫印迹法(WB)检测TRIM44、IKKβ、IκBα、磷酸化核因子-κB抑制蛋白α(p-IκBα)、NF-κB p65、B淋巴细胞瘤-2(Bcl-2)、基质金属蛋白酶-9(MMP-9)、半胱氨酸天冬氨酸蛋白酶3(Caspase3)、血管生成因子(VEGF)蛋白表达差异.结果:与空白组相比,顺铂组、中药组、联合组均可抑制OC裸鼠移植瘤的生长;各用药组均能够降低瘤体组织中IKKβ、NF-κB p65mRNA表达,升高IκBα mRNA表达,且联合组差异最为显著(P<0.05);各用药组均能显著降低瘤体组织中TRIM44、IKKβ、p-IκBα、NF-κB p65、Bcl-2、MMP-9、VEGF蛋白表达,升高IκBα、Caspase-3蛋白表达,且以联合组差异为著(P<0.05).结论:理冲生髓饮能够抑制OC裸鼠移植瘤的生长,并协同顺铂增强抗肿瘤的作用.其作用机制可能是通过下调TRIM44表达,抑制NF-κB信号通路的激活,从而抑制OC侵袭、转移,促进其凋亡.

Objective:To explore the relevant mo-lecular mechanism of Lichong Shengsui Decoction(理冲生髓饮)exerting anti-ovarian cancer(OC)effects by regulating the nuclear factor-κB(NF-κB)pathway mediated by tripartite motif-containing protein 44(TRIM44).Methods:Human ovarian cancer SKOV3 cells with stable overexpression of TRIM44 were constructed,and the cells were subcutaneously inoculated into Balb/C nude mice to establish a xenograft tumor model.A total of 40 nude mice were randomly divided into the blank group,cisplatin group,Chinese medicinal group,and combination group,with 10 mice in each group.The Chinese medicinal group was given Lichong Shengsui Decoction at a dose of 7.52 g/kg by gavage twice a day.The cisplatin group was intraperitoneally injected with 0.1 mg/mL cisplatin solution every other day.The combination group was given Lichong Shengsui Decoction at 7.52 g/kg by gavage twice a day,combined with intraperitoneal injection of 0.1 mg/mL cisplatin solution(equivalent to 3 mg/kg)every other day.The blank group was given an equal volume of normal saline by gavage twice a day.After 14 days of drug intervention,the tumor volume and growth trend of nude mice were observed.Hematoxylin-eosin(HE)staining was used to observe the histopathological changes of tumor tissues.Reverse transcription-polymerase chain reaction(RT-PCR)was employed to detect the mRNA expressions of inhibitor of κB kinase β(IκKβ),nuclear factor-κB inhibitorα(IκBα),and nuclear factor-κB p65(NF-κB p65)in tumor tissues.Double immunofluorescence staining was used to detect the distribution of TRIM44 and NF-κB p65 proteins in tumor tissues.Western blot(WB)was applied to determine the differences in the protein expressions of TRIM44,IκKβ,IκBα,phosphorylated nuclear factor-κB inhibitor α(p-IκBα),NF-κB p65,B-cell lymphoma-2(Bcl-2),matrix metalloproteinase-9(MMP-9),cysteinyl aspartate-specific protease 3(Caspase3),and vascular endothelial growth factor(VEGF)in tumor tissues.Results:Compared with the blank group,the cisplatin group,Chinese medicinal group,and combination group all inhibited the growth of ovarian cancer xenografts in nude mice.All medication groups were able to reduce the mRNA expressions of IκKβ and NF-κB p65 in tumor tissue,and up-regulate the mRNA expression of IκBα in tumor tissues,with the most significant difference in the combination group(P<0.05).All medication groups significantly decreased the protein expressions of TRIM44,IκKβ,p-IκBα,NF-κB p65,Bcl-2,MMP-9,VEGF,and increased the protein expressions of IκBαand Caspase-3 in tumor tissues,with the most prominent effect in the combination group(P<0.05).Conclusions:Lichong Shengsui Decoction can inhibit the growth of ovarian cancer xenografts in nude mice and enhance anti-tumor effects in combination with cisplatin.Its mechanism may be related to down-regulating the expression of TRIM44,inhibiting the activation of the NF-κB signaling pathway,thereby suppressing the invasion and metastasis of ovarian cancer and promoting its apoptosis.
