通过探讨抑菌剂浓度和激素的配组对耐寒月季离体快繁的影响,建立耐寒月季"2021-MH"开放式组培快繁体系,为耐寒月季工厂化种苗生产提供技术支撑.以耐寒月季新品系"2021-MH"为材料,在组培快繁的各个阶段(初代培养、继代培养、生根培养),添加不同浓度组合的抑菌剂(ZP抗菌素、特美汀、PPMTM)及植物生长激素,综合分析抑菌剂和激素组合对耐寒月季开放式培养的效果.结果表明:初代培养阶段最佳的培养基抑菌剂和激素浓度配组(抑菌剂:60mg•L-1 ZP,100mL•L-1 特美汀,1.0mL•L-1 PPMTM;培养基:MS 加1.5mg•L-16-BA加 0.1mg•L-12,4-D加 0.3mg•L-1 IBA),出芽率为 96.15%,污染率为 13.33%,褐化率为3.85%;继代培养阶段最佳的抑菌剂和激素浓度配组(抑菌剂:50mg•L-1 ZP,150mL•L-1 特美汀,0.75mL•L-1 PPMTM;培养基:MS加 2.5mg•L-1 6-BA加1.0mg•L-1 2,4-D加0.2mg•L-1 NAA),污染率为5.00%,增殖系数为 5.43,玻璃化率为 1.75%;生根培养阶段最佳的培养基抑菌剂和激素浓度配组,(抑菌剂:30mg•L-1 ZP,50mL•L-1特美汀,0.75mL•L-1 PPMTM;培养基:1/2MS 加 1.0mg•L-1 IAA 加 0.5mg•L-1 NAA),污染率为10.00%,生根率为 94.44%,幼苗生根快,茎粗,叶色浓绿.成功建立了耐寒月季"2021-MH"开放式组培快繁体系,该体系具有污染率低,不定芽增殖系数高,褐化率、玻璃化率低,幼苗生长状态佳等优点.

By discussing the effects of bacteriostatic concentration and hormone combination on in vitro rapid propagation of cold-resistant rose,an open tissue culture rapid propagation system of cold-resistant rose"2021-MH"was established to provide technical support for industrialized seedling production of cold-resistant rose.A new cold-tolerant rose strain"2021-MH"was used as the material.At each stage of tissue culture and rapid propagation(primary culture,subculture and rooting culture),different concentration combinations(ZP antibiotics,Timentin and PPMTM)and plant growth hormones were added,and the effects of bacteriostatic agents and hormone combinations on open culture of cold-tolerant rose were comprehensively analyzed.The results indicate:The best combination of bacteriostatic agent and hormone concentration in the primary culture stage(bacteriostatic agent:60mg•L-1 ZP,100mL•L-1 Timentin,1.0mL•L-1 PPMTM;Medium:MS+1.5mg•L-1 6-BA+0.1mg•L-1 2,4-D+0.3mg•L-1 IBA),the germination rate was 96.15%,the contamination rate was 13.33%,the browning rate was 3.85%;The best combination of bacteriostatic agent and hormone concentration in subculture stage(bacteriostatic agent:50mg•L-1 ZP,150mL•L-1 Timentin,0.75mL•L-1 PPMTM;Medium:MS+2.5mg•L-1 6-BA+1.0mg•L-1 2,4-D+0.2mg•L-1 NAA),with pollution rate of 5.00%,proliferation coefficient of 5.43,vitrification rate of 1.75%;The best combination of culture medium bacteriostatic agent and hormone concentration in rooting culture stage(bacteriostatic agent:30mg•L-1 ZP,50mL•L-1 Timentin,0.75mL•L-1 PPMTM;Medium:1/2MS+1.0mg•L-1 IAA+0.5mg•L-1 NAA),the contamination rate was 10.00%,the rooting rate was 94.44%,the seedlings took root quickly,the stems were thick and the leaves were dark green.An open tissue culture and rapid propagation system for the cold-tolerant rose"2021-MH"was successfully established,which has the advantages of low contamination rate,high adventitious bud proliferation coefficient,low browning rate and vitrification rate,and favorable seedling growth performance.
