Effects of Different Breeding Environments on Animal Immunological Homeostasis and Immune Response

为探讨不同选育环境对动物免疫系统发育与功能调控的影响,选取源自同一群体且在无特定病原体环境(SPF)与普通环境(P)下,经过10年选育的2组新西兰兔,各24只,通过血液生理生化指标检测、LPS刺激试验及TLR4信号通路相关基因表达分析,比较其免疫稳态与应答差异.结果表明:P兔的白细胞数、中性粒细胞数、红细胞数、肌酸激酶等血液生理生化指标均显著高于SPF兔(P<0.05),表明其免疫系统在自然微生物暴露下维持较高活性.LPS刺激后,P兔在基因表达层面表现出迅速且强烈的免疫应答,TLR4及其下游MyD88、NF-κB等关键信号分子显著上调,与基因表达的快速变化相比,多项免疫指标变化相对平缓,提示其免疫反应以"快速信号启动、稳健效应输出"为特征;SPF培育兔启动明显滞后且幅度较弱,在体液层面的波动更大.普通环境培育兔显示出成熟免疫系统特有的精细调控能力,SPF培育兔的免疫系统仍处于静默化状态,缺乏有效的稳态调节机制.长期不同环境选育塑造了动物的免疫差异性:适度的环境微生物暴露有助于维持免疫系统的稳态和应答能力,而长期高洁净环境可能导致其免疫敏感性下降.研究结果揭示了不同免疫环境在动物免疫适应性形成中的关键作用,印证了"卫生假说"在集约化养殖中的警示意义,为动物遗传选育与健康养殖体系优化奠定了理论依据.

To investigate the effects of different breeding environments on the development and functional regulation of the animal immune system,this study selected two groups of New Zealand rabbits(24 each)originating from the same population but bred for 10 years in either a specific pathogen-free(SPF)or a conventional environment(P).Immune homeostasis and response were compared through blood physiological and biochemical analysis,LPS challenge tests,and gene expression analysis of the TLR4 signaling pathway.The results showed that P rabbits had significantly higher physiological and biochemical indicators,including total white blood cell count,neutrophil percentage,red blood cell count and creatine kinase,than SPF rabbits(P<0.05),indicating maintained higher immune activity under natural microbial exposure.Following LPS stimulation,rabbits bred in the conventional environment exhibited an exceptionally rapid and intense immune response at the gene expression level,with marked upregulation of key signaling molecules such as TLR4 and its downstream effectors of MyD88 and NF-κB.In contrast to the swift alterations in gene expression,the changes in multiple immune parameters were relatively moderate,suggesting that their immune reaction was characterized by"rapid signal initiation with robust effector output".In comparison,SPF-bred rabbits demonstrated a notably delayed and weaker response initiation,accompanied by more pronounced fluctuations in humoral parameters.Thus,rabbits raised in conventional conditions displayed the fine-tuned regulatory capacity typical of a mature immune system,whereas the immune system of SPF-bred rabbits remained in a naive state,lacking effective homeostatic regulatory mechanisms.The findings demonstrate that long-term breeding in distinct environments shapes immune divergence:Moderate microbial exposure helps maintain immune homeostasis and responsiveness,whereas prolonged high cleanliness may lead to reduced immune sensitivity.This study reveals the critical role of different immune environments in shaping immune adaptability,confirms the warning significance of the hygiene hypothesis in intensive farming,and provides a theoretical basis for optimizing animal genetic breeding and healthful farming systems.
