柴芪益肝颗粒通过抑制PI3K/AKT和ERK信号通路延缓肝细胞癌进展OA
Chaiqi Yigan Granules Delay Progression of Hepatocellular Carcinoma by Inhibiting PI3K/AKT and ERK Signaling Pathways
目的 探讨柴芪益肝颗粒对H22肝癌荷瘤小鼠的作用及机制.方法 50只SPF级雄性昆明小鼠,采用H22肝癌细胞左腋下皮下注射构建H22肝癌荷瘤小鼠模型,模型构建成功后,采用随机数字表法将小鼠随机分为模型组、柴芪益肝颗粒低剂量组、柴芪益肝颗粒中剂量组、柴芪益肝颗粒高剂量组及索拉非尼组,每组10只,按照分组灌胃给药14 d后取材.HE染色观察小鼠肿瘤组织病理形态变化,TUNEL染色观察肿瘤组织细胞凋亡程度;RT-qPCR法检测肿瘤组织凋亡相关基因B淋巴细胞瘤2(Bcl-2)、Bcl-2相关x蛋白(Bax)、活化半胱氨酸蛋白酶3(Caspase-3)的mRNA表达;Western blot法检测小鼠肿瘤组织凋亡相关蛋白Bel-2、Bax、Caspase-3,侵袭及迁移蛋白N-cadherin、MMP7、Vimen-tin和E-cadherin,以及PI3K/AKT/ERK信号通路的蛋白表达;免疫组化法检测Caspase-3在肿瘤组织的表达.结果 与模型组相比,柴芪益肝颗粒可以有效延缓肿瘤进展,促进肿瘤细胞的凋亡,上调肿瘤组织Bax、Caspase-3的mRNA及蛋白表达,下调肿瘤组织Bel-2的mRNA及蛋白表达,增强肿瘤组织E-cadherin的蛋白表达,降低肿瘤组织中N-cad-herin、MMP7 和Vimentin蛋白的表达,以及抑制PI3K/AKT和ERK信号通路的蛋白表达.结论 柴芪益肝颗粒可能通过抑制PI3K/AKT和ERK信号通路促进肿瘤细胞凋亡,抑制其上皮-间质转化,从而延缓肝癌进展.
Objective To investigate the effect and mechanism of Chaiqi Yigan Granules(柴芪益肝颗粒)on H22 hepatocel-lular carcinoma bearing mice.Methods Fifty specific pathogen-free male Kunming mice were used to establish H22 hepatoma tumor-bearing mouse models by subcutaneous injection of H22 hepatoma cells into the left armpit.After the models were suc-cessfully constructed,the mice were randomly divided into model group,Chaiqi Yigan Granules low-dose group,Chaiqi Yigan Granules medium-dose group,Chaiqi Yigan Granules high-dose group and sorafenib group by random number table method(10 per group).The samples were collected 14 days after intragastric administration according to the group.HE staining was used to observe the pathological morphological changes of tumor tissues.TUNEL staining was used to observe the degree of apoptosis in tumor tissues.RT-qPCR was used to detect the mRNA expressions of apoptosis-related genes B-cell lymphoma 2(Bel-2),Bel-2 associated X protein(Bax)and activated cysteine proteinase 3(Caspase-3).The expressions of apoptosis-related pro-teins Bel-2,Bax,Caspase-3,invasion and migration proteins N-cadherin,MMP7,Vimentin,E-cadherin and PI3K/AKT/ERK signaling pathway were detected by Western blot.The expression of Caspase-3 in tumor tissues was detected by immunohis-tochemistry method.Results Compared with the model group,Chaiqi Yigan Granules could effectively delay the progression of tumor,promote the apoptosis of tumor cells,up-regulated mRNA and protein expressions of Bax and Caspase-3,down-regula-ted mRNA and protein expressions of Bel-2,enhanced the protein expression of E-cadherin,decreased the protein expressions of N-cadherin,MMP7 and Vimentin in tumor tissue and inhibited the protein expression of PI3K/AKT/ERK signaling pathway.Conclusion Chaiqi Yigan Granules may promote tumor cell apoptosis and inhibit the epithelial-mesenchymal transition by inhib-iting PI3K/AKT and ERK signaling pathways,thus delaying the progression of hepatocellular carcinoma.
