桑叶总生物碱提取及其与牛血清蛋白结合能力研究OA
Extraction of Total Alkaloids From Mulberry Leaves and Its Binding With Bovine Serum Albumin
采用超声波低温提取工艺从桑叶中提取总生物碱,在单因素试验的基础上通过响应面法优化提取工艺,利用液相色谱/质谱联用仪对桑叶总生物碱主要结构进行鉴定分析,并通过系列光谱研究了桑叶总生物碱与牛血清蛋白(BSA)的结合能力.桑叶总生物碱较佳提取工艺为桑叶3.0 g、0.05 mol/L盐酸溶液与桑叶的液料比10∶1(mL∶g)、再加入乙醇配制成乙醇体积分数33%的混合溶液,在超声波功率250 W、提取温度40℃、提取时间30 min、提取2 次.在此条件下桑叶总生物碱提取量为2.596±0.13 mg/g.经液相色谱/质谱分析可知,桑叶总生物碱主要有4 种结构:2-氧-α-D-半乳吡喃糖苷-脱氧野尻霉素、1-脱氧野尻霉素(1-DNJ)、1,4-二脱氧-1,4-亚氨基-D-阿拉伯糖醇与荞麦碱.由荧光光谱分析可知,桑叶总生物碱能够与牛血清蛋白结合,结合常数为(9.79±0.23)×103 L/(mol·s).圆二色光谱分析结果证实了桑叶总生物碱可以改变牛血清蛋白二级结构.
The total alkaloids were extracted from mulberry leaves by ultrasonic low-temperature extraction process.Based on single factor experiments,the extraction process was optimized by response surface method.The main structures of total alkaloids from mulberry leaves were identified and analyzed by liquid chromatography/mass spectrometry,and the binding ability of total alkaloids from mulberry leaves to bovine serum albumin(BSA)was studied through a series of spectra.The results showed that the optimal extraction process of total alkaloids from mulberry leaves was as follows:mulberry leaves of 3.0 g,solid-liquid ratio of 0.05 mol/L hydrochloric acid solution to mulberry leaves10∶1(mL∶g),ethanol volume fraction of 33%,ultrasonic power of 250 W,extraction temperature of 40℃,extraction time of 30 min,and two times of extraction.Under these conditions,the extraction amount of total alkaloids from mulberry leaves was 2.596±0.13 mg/g.According to the analysis by liquid chromatography/mass spectrometry(LC/MS),there were four main structures of total alkaloids from mulberry leaves,2-O-α-D-galactopyranosyl-deoxynojirimycin,1-deoxynojirimycin(1-DNJ),1,4-dideoxy-1,4-imino-D-arabinitol and fagomine.It could be seen from the fluorescence spectrum that the total alkaloids from mulberry leaves could bind to BSA with a binding constant of(9.79±0.23)×103 L/(mol·s).Circular dichroism analysis confirmed that total alkaloids from mulberry leaves could alter the secondary structure of BSA.
