miR-222作为在高脂和低脂中国荷斯坦奶牛中表达有显著性差异的miRNAs,其在牛乳腺上皮细胞(bMECs)中的功能尚不清楚.为明确miR-222对奶牛乳腺上皮细胞乳脂代谢的影响,通过将miR-222的过表达和干扰载体分别转染进bMECs中,检测bMECs内甘油三酯、胆固醇含量,同时对bMECs的增殖和凋亡水平进行检测.结果表明:与miR-shNC组相比,过表达miR-222可显著降低细胞内甘油三酯含量(P<0.05),降低胞内胆固醇含量(P<0.05);显著促进脂肪酸代谢基因PPARA,ELOVL6,ACSL1和FADS1的mRNA表达(P<0.05);显著抑制乳腺上皮细胞生长,并促进其凋亡(P<0.05).说明miR-222不仅参与调控bMECs的凋亡,还参与调控bMECs的乳脂合成以及脂肪酸代谢过程,该研究为探讨miR-222在bMECs乳脂代谢中的调控作用和分子机制奠定基础.

The function of miR-222 in bovine mammary epithelial cells(bMECs),as a miRNAs dif-ferentially expressed in high-fat and low-fat Chinese Holstein cows,is still unclear.Therefore,this study aims to verify the effects of miR-222 on milk fat metabolism in cow mammary epithelial cells,and improve the molecular mechanism of miRNA regulating milk fat synthesis in cows.The overex-pression and interference vectors of miR-222 were transfected into bMECs respectively to detect the contents of triglycerides and cholesterol in bMECs.At the same time,the proliferation and apoptosis of bMECs were detected.The results showed that overexpression of miR-222 could significantly re-duce the contents of triglycerides in cells(P<0.05)and intracellular cholesterol(P<0.05)compared with miR-shNC group.It significantly promoted the mRNA expression of PPARA,ELOVL6,ACSL1,and FADS1 genes involved in fatty acid metabolism.It can significantly inhibit the growth of mam-mary epithelial cells and promote their apoptosis.The results showed that miR-222 was not only in-volved in regulating the fate of bMECs,but also involved in regulating the milk fat synthesis and fatty acid metabolism of bMECs,laying a foundation for further exploring the regulatory role and molecu-lar mechanism of miR-222 in the milk fat metabolism of bMECs significantly correlated with lactose status(P<0.05).The results of this study can provide a basis for molecular breeding of Chinese Hol-stein dairy cows and further improving the quality and performance of dairy products.
