菜豆原生质体的融合条件研究OA
Fusion Conditions of Protoplast from Common Bean
原生质体融合是创制、选育严格自花授粉作物新品种的有效手段,聚乙二醇(PEG)-高Ca2+-高pH融合法是高效促进原生质体融合的理想方法.为建立菜豆原生质体融合体系,利用PEG-高Ca2+-高pH法诱导菜豆原生质体融合,研究菜豆原生质体适宜的融合方法和条件.结果表明:菜豆原生质体融合的适宜条件为菜豆原生质体密度1.3×105/mL,先用35%PEG(MW=6 000)诱导融合,然后连续4次加入20 µL高Ca2+-高pH溶液,常温下保温60~90 min,最后用80 µL的高Ca2+-高pH溶液洗涤以促进融合,连续洗涤2~3次,融合率可达13%.
Protoplast fusion is an effective means to create and breed new varieties of strictly self-pollinated crops.PEG-high Ca2+-high pH fusion method is an ideal method to promote protoplast fu-sion with high efficiency.To establish a protoplast fusion system for common bean,the protoplast fu-sion of common bean was induced by PEG-high Ca2+-high pH method,and the suitable fusion meth-ods and conditions for bean protoplasts were studied.The results showed that the optimal conditions for protoplast fusion in beans are as follows:a protoplast density of 1.3×105/mL,initial induction of fusion with 35%PEG(MW=6 000),followed by the sequential addition of 20 µL of high Ca2+-high pH solution four times,incubation at room temperature for 60-90 min,and finally washing to pro-mote fusion with with 80 µL of high Ca2+-high pH solution for 2-3 times,whcih can achieve a fusion rate of up to 13%.
