β-胡萝卜素干预对育成鸡免疫损伤的缓解作用OA
Alleviating Effect of β-Carotene Intervention on Immune Damage of Growing Chickens
为探究β-胡萝卜素干预对育成鸡免疫损伤的影响,选取1日龄海兰褐鸡180只,分为空白对照组和β-胡萝卜素组,饲喂至65日龄时进行二次分组,分别腹腔注射0,125,150 mg/kg环磷酰胺(Cy),注射1周后采样检测.结果表明:与对照组相比,β-胡萝卜素干预能降低免疫损伤鸡血液中促炎因子IL-2,IFN-γ,TNF-α的含量,增加免疫损伤鸡肠道绒毛长、降低隐窝深、增加肠道绒隐比;显著增加免疫损伤鸡肠道中Claudin-1,Occuldin,ZO-1,IgA mRNA相对表达量(P<0.05);显著降低肝脏、脾脏、胸腺、法氏囊中CHOP,TNF-α,Caspase-3,IL-1β mRNA相对表达量(P<0.05),增加Bcl-2和Nrf2 mRNA相对表达量(P<0.05).说明β-胡萝卜素能通过增加免疫损伤鸡的器官重,修复肠道黏膜屏障,降低细胞凋亡和氧化应激相关基因mRNA的表达量以及血液中促炎因子含量,来发挥其对育成鸡免疫损伤的缓解作用.研究结果为预防性减缓育成鸡免疫损伤,以及鸡健康养殖中功能性饲料添加剂的利用提供参考.
In order to explore the effect of β-carotene intervention on immune damage in the process of chicken breeding,180 1-day-old Hy-line brown chickens were divided into control group and β-carotene group,and when they were fed to 65 days of age,they were injected with 0,125 and 150 mg/kg Cyclophosphamide(Cy),and sampled for detection one week after injection.The results showed that,compared with the control group,β-carotene intervention could reduce the contents of pro-inflammatory factors IL-2,IFN-γ and TNF-α in the blood of immune-damaged chickens,increase the length of intestinal villi,reduce the depth of crypts and increase the ratio of intestinal villi to crypts in immune-damaged chickens,significantly increase the relative mRNA expression levels of Claudin-1,Occuldin,ZO-1 and IgA in the intestinal tract of immune-damaged chickens(P<0.05),significantly down-regulate liver,spleen,thymus.The relative mRNA expressions of CHOP,TNF-α,Caspase-3 and IL-1β in the bursa were decreased(P<0.05),and the relative mRNA expressions of Bcl-2 and Nrf2 were increased(P<0.05).β-carotene intervention can increase the weight of immune-damaged chickens,repair the intestinal mucosal barrier,reduce the mRNA expression of apoptosis and oxidative stress-related genes,and reduce the content of pro-inflammatory factors in the blood,so as to play a role in alleviating immune injury.The results of this study provide a preliminary exploration for the preventive alleviation of immune damage in finishing chickens,and provide a reference for the full utilization of functional feed additives in healthy breeding.
