为探究过氧化物酶增殖物激活受体γ(Peroxisome proliferator-activated receptors γ,PPARγ)在性成熟前公羊生殖器官中的表达情况,以性成熟前雄性绵羊为研究对象,测量睾丸、附睾和输精管等表型参数,采用免疫组织化学染色、实时荧光定量PCR和免疫印迹法,检测PPARγ在性成熟前公羊生殖器官中的表达及定位.结果表明:大睾丸组睾丸质量、纵轴长、输精管长、横轴长和附睾质量均显著高于小睾丸组(P<0.05);PPARγ蛋白在睾丸支持细胞、间质细胞、各级生精细胞,以及附睾和输精管的上皮细胞、精子、纤毛中均有表达;大睾丸组PPARγ mRNA和蛋白在睾丸、输精管、附睾头、附睾体、附睾尾中的相对表达量均高于小睾丸组.推测PPARγ可能与睾丸发育有关,是影响绵羊繁殖性能的关键因子,研究结果为揭示精子发生发育过程提供了新思路.

In order to explore the expression of peroxidase proliferator-activated receptor γ(Peroxisome proliferator-activated receptors γ,PPARγ)in the reproductive organs of rams before sexual maturity,prepubertal male sheep were used as the research subjects.And the parameters of testes,epididymis and vas deferens were measured.Immunohistochemistry,fluorescence quantitative PCR and immunoblotting were used to detect the expression and localization of PPARγ in the reproductive organs of the male sheep before sexual maturity.The results showed that the testicular weight,longitudinal axis length,vas deferens length,transverse axis length and epididymal weight in the large testicular group were significantly higher than those in the small testicular group(P<0.05).PPARγ protein was expressed in sertoli cells,mesenchymal cells,spermatogenic cells at all levels,and epididymis and vas deferens of epidimelial cells,spermatozoa and cilia.The expression levels of PPARγ mRNA and protein in the testis,vas deferens,caput epididymidis,corpus epididymidis and cauda epididymidis in the large testis group were higher than those in the small testis group.It is speculated that PPARγ may be related to testicular development and is a key factor affecting the reproductive performance of sheep,and provide a new path for revealing the process of spermatogenesis and development.
