基因2型鹅星状病毒(Goose astrovirus genotype 2,GAstV-2)是引发雏鹅痛风的主要病原体,基于2022年对GAstV-2山东分离株的分析结果,对2023年分离的2株GAstV的基因序列进行测定及遗传进化分析.结果表明:分离的2株GAstV均为GAstV-2 Ⅱ-c亚型,与参考株一致,表明Ⅱ-c亚型是我国当前流行的优势基因型.进化分析结果表明,G666和G667与山东分离株G529,G538,G548,G576的亲缘关系较近,在氨基酸序列中出现多处一致的规律性和特征性突变,在呈现新的遗传标记(ORF1b 第483位,ORF2第228位和376位)的同时,"温度响应型"位点已向适应高温的类型突变,表明病毒在不断进化.对GAstV-2的遗传进化进行分析,研究结果揭示了病毒的流行状况和演化规律,为基因2型鹅星状病毒疫苗和诊断试剂的研发奠定基础.

Goose astrovirus genotype 2(GAstV-2)is considered as the major agent of fatal visceral gout in goslings.In this study,based on the GAstV-2 isolated in Shandong Province previously,2 strains of GAstV-2 were further isolated during specific period in 2023.Both viral full-length genomes were sequenced and genetic evolutionary analysis was conducted.Our results indicated that the strains isolated in this study were both GAstV-2 subgenotype Ⅱ-c,indicating that subgenotypeⅡ-c was still the dominant genotype in China.Phylogenetic analysis of genome and three open reading frames demonstrated that G666 and G667 were clustered with G529,G538,G548 and G576,which were isolated in Shandong.Moreover,several regular and characteristic mutations were observed in the six strains.The mutations contained three novel genetic markers(positions 483 in ORF1b,and position 228 and 376 in ORF2)and six"temperature response"residues suggested in previous study,demonstrating viral durative evolution.Overall,these findings improved our understanding for epidemiology and genetic evolution of GAstV-2 and contributed to the development of vaccines and diagnostics.
