猪流行性腹泻病毒结构蛋白对细胞极性分子Par6表达的影响OA
Effects of Porcine Epidemic Diarrhea Virus Structural Proteins on the Expression of Cellular Polar Molecule Par6
紧密连接(Tight junction,TJ)是肠上皮细胞间的主要连接结构,能抵御外界病原体侵袭,还可以选择性地调节小分子物质和离子进入体内.在肠上皮细胞中,极性复合物能与紧密连接相互作用,维持细胞极性.为研究猪流行性腹泻病毒(Porcine epidemic diarrhea virus,PEDV)结构蛋白对细胞极性分子Par6表达的影响,构建了携带PEDV N,S1,S2,M,E基因的真核表达质粒,利用Lipofectamine 3000转染试剂,分别设置转染时间组和转染剂量组,将真核表达质粒转染至猪小肠上皮细胞(IPEC-J2),其中,时间组选取5 μL并在转染24,36,48 h收集样品,剂量组选取1,3,5 μL并在转染24 h收集样品,检测细胞中Par6 mRNA和蛋白的表达量.结果表明:转染携带PEDV N,S1,S2,M和E蛋白的真核表达质粒均可上调细胞中极性分子Par6 mRNA和蛋白的表达量,且N蛋白的上调作用最显著.研究结果为揭示PEDV致病机制提供参考.
Tight junctions(TJs)are the major junctional structures among intestinal epithelial cells,which can resist the invasion of external pathogens and selectively regulate the entry of small molecules and ions into the body.In intestinal epithelial cells,polarity complexes can interact with tight junctions to maintain cell polarity.To investigate the effects of porcine epidemic diarrhea virus(PEDV)structural proteins on the expression of the cell polarity molecule Par6,eukaryotic expression plasmids carrying PEDV N,S1,S2,M and E genes were constructed.Using Lipofectamine 3000 transfection reagent,time-course and dose-response groups were set up for transfection into porcine jejunum epithelial cell line(IPEC-J2).Specifically,in the time-course group,cells were transfected with a fixed dose of 5 μL and samples were collected at 24,36 and 48 hours post-transfection;in the dose-response group,cells were transfected with 1,3 and 5 μL of the reagent and samples were collected at 24 hours post-transfection.The expression levels of Par6 mRNA and protein in the cells were then detected.The results showed that transfection with eukaryotic expression plasmids carrying PEDV N,S1,S2,M and E proteins all up-regulated the expression of Par6 mRNA and protein in the cells,with the N protein exerting the most significant up-regulatory effect.This study provides a reference for revealing the pathogenic mechanism of PEDV.
