中医药靶向调控线粒体质量控制系统治疗脑缺血再灌注损伤研究进展OA
Progress of Chinese medicine targeting and regulating mitochondrial quality control system for the treatment of cerebral ischemia/reperfusion injury
脑缺血再灌注损伤(cerebral ischemia-reperfusion injury,CIRI)是指脑组织在缺血后恢复血流灌注时可能引发的继发性病理损害.线粒体是细胞的"能量动力室",具有供应能量、代谢活性氧、储存钙以及介导细胞程序性死亡等重要生理功能.因此维持线粒体稳态对神经细胞的生存及功能恢复具有决定性作用.而线粒体质量控制系统(mitochondrial quality control,MQC)通过调控线粒体动力学、线粒体自噬、线粒体生物发生和线粒体转移等过程,可减轻CIRI所致的线粒体结构和功能损伤.近年来,中医药疗法应用较为广泛,可通过多途径、多靶点、多层次明显改善CIRI.因此就MQC在CIRI中的作用机制以及中医药通过调控MQC在CIRI中的研究进展进行综述,为今后治疗CIRI提供新的思路和研究方向.
Cerebral ischemia-reperfusion injury(CIRI)is defined as secondary pathological damage that may be triggered when blood perfusion is restored to brain tissue after ischemia.Mitochondria are the"energy powerhouse"of the cell,with impor-tant physiological functions such as supplying energy,metabolizing reactive oxygen species,storing calcium,and mediating pro-grammed cell death.Maintaining mitochondrial homeostasis is therefore decisive for the survival and functional recovery of nerve cells.In contrast,the mitochondrial quality control(MQC)can attenuate CIRI-induced mitochondrial structural and functional damage by regulating the processes of mitochondrial dynamics,mitochondrial autophagy,mitochondrial biogenesis,and mitochon-drial translocation.In recent years,TCM therapies have been more widely applied,which can significantly improve CIRI through multiple pathways,targets and levels.Therefore,a review on the mechanism of MQC in CIRI and the research progress of TCM in CIRI by modulating MQC is conducted to provide new ideas and research directions for the treatment of CIRI in the future.
