为建立甘蔗梢多酚类化合物的高效检测方法,本研究采用高效液相色谱-质谱联用技术(HPLC-MS),对甘蔗梢中多酚成分进行系统定性、定量分析.样品经复合酶辅助提取(复合酶用量1.6%、pH 5.0、温度 56℃、时间 30 min),并通过大孔树脂柱纯化,最终以 0.22 μm微孔滤膜过滤后进样.结果表明,甘蔗梢提取物中共鉴定出 8 种多酚单体,包括奎尼酸、安息香酸、咖啡酸、芹菜素、对香豆酸、绿原酸、阿魏酸和没食子酸.其中,对香豆酸、阿魏酸、绿原酸和没食子酸在样品中普遍存在,含量分布于 1.8~30 mg/g之间.研究进一步对这 4 个组分进行了定量方法学验证,显示其线性关系良好(相关系数R2>0.99),加标回收率介于 90.78%~101.06%之间,精密度试验的相对标准偏差(RSD)为 0.26%~2.24%(n=6),方法检出限为 0.1~0.4 mg/kg,定量限为 0.5~2.1 mg/kg.该方法分离效果好、灵敏度高、重复性佳,适用于甘蔗梢多酚的快速定性、定量分析,为其在功能食品与药品领域的进一步开发利用提供了可靠的分析依据.

To establish an efficient detection method for polyphenolic compounds in sugarcane tops,this study employed high performance liquid chromatography-mass spectrometry(HPLC-MS)for systematic qualitative and quantitative analysis of polyphenols in sugarcane tops.The samples were extracted via composite enzyme-assisted extraction(1.6%composite enzyme,pH 5.0,56℃,30 min),purified by macroporous resin column chromatography,and finally filtered through a 0.22 μm microporous membrane before injection.The results showed that eight polyphenol monomers were identified in the sugarcane top extract,including quinic acid,benzoic acid,caffeic acid,apigenin,p-coumaric acid,chlorogenic acid,ferulic acid,and gallic acid.Among these,p-coumaric acid,ferulic acid,chlorogenic acid,and gallic acid were commonly present in the samples,with concentrations ranging from 1.8 to 30 mg/g.Further methodological validation for the quantification of these four components demonstrated good linearity(correlation coefficient R2>0.99),spike recovery rates between 90.78%and 101.06%,relative standard deviation(RSD)for precision tests ranging from 0.26%to 2.24%(n=6),method detection limits of 0.1~0.4 mg/kg,and quantification limits of 0.5~2.1 mg/kg.This method exhibits excellent separation efficiency,high sensitivity,and good reproducibility,making it suitable for the rapid qualitative and quantitative analysis of polyphenols in sugarcane tops.It provides a reliable analytical foundation for further development and utilization of sugarcane tops in functional foods and pharmaceuticals.
