[目的]对猪circTEX9的生物学特性进行鉴定,并探讨猪circTEX9对猪肌内脂肪沉积的调控作用.[方法]以前期测序数据筛选到的circTEX9为研究对象,采用Sanger测序和亚细胞定位等方法,对其环状结构、稳定性和亚细胞定位等生物学特性进行鉴定和分析;同时过表达circTEX9探究其对猪肌内前体脂肪细胞增殖与分化的调控作用,结合生物学信息方法预测其调控的下游靶基因及其参与的信号通路.[结果]猪circTEX9是由TEX9基因的外显子7~10构成的环状RNA分子,具有RNase R抗性,主要定位于细胞质中.circTEX9的二级结构稳定,且包含2个开放阅读框和2个内部核糖体进入位点,有翻译潜能.circTEX9显著抑制增殖标志基因的表达水平,且显著降低细胞活力,促进分化标志基因的表达水平.基于上述结果和生物信息学分析,预测circTEX9通过竞争性内源RNA(competing endogenous RNA,ceRNA)网络吸附ssc-miR-204、ssc-miR-23a和ssc-miR-23b,进而调控下游靶基因.下游靶基因通过参与单磷酸腺苷激活的蛋白激酶信号通路、钙信号通路和丝裂原活化蛋白激酶信号通路等发挥生物学功能,对猪肌内前体脂肪细胞产生影响.[结论]circTEX9在猪体内稳定性较高,具有抑制肌内前体脂肪细胞的增殖并促进脂质沉积的作用,可通过ceRNA网络发挥其生物学功能.

[Objective]The biological characteristics of porcine circTEX9 were identified and the regu-lation role of porcine circTEX9 on intramuscular fat deposition was studied.[Method]Based on the previous sequencing data of our research group,circTEX9 was selected as the research object,and its ring structure,stability and subcellular localization were comprehensively identified and analyzed by Sanger sequencing and subcellular localization.At the same time,the regulatory effects of circTEX9 on the proliferation and differentiation of porcine intramuscular preadipocytes were investigated by overexpression method,and the downstream target genes regulated by circTEX9 and the signal path-ways involved were predicted by combining biological information methods.[Result]Porcine circ-TEX9 is a circular RNA molecule composed of exons 7 to 10 of TEX9 gene,which is resistant to RNase R and mainly exists in the cytoplasm.circTEX9 has a stable secondary structure and contains two open reading frames(ORF)and two internal ribosome entry sites(IRES)with translation poten-tial.circTEX9 significantly inhibited the expression levels of proliferative marker genes,and signifi-cantly decreased cell viability.The expression levels of differentiation marker genes were significantly upregulated.Based on the above results and bioinformatics analysis,it was predicted that circTEX9 could absorb ssc-miR-204,ssc-miR-23a and ssc-miR-23b through competing endogenous RNA(ceRNA)network to regulate downstream target genes.These target genes exert biological functions by participating in the adenosine 5 ′-monophosphate-activated protein kinase signaling pathway,cal-cium signaling pathway and mitogen-activated protein kinase signaling pathway,ultimately influencing porcine intramuscular preadipocytes.[Conclusion]circTEX9 has high stability in pig vivo and inhibits the proliferation of intramuscular preadipocytes while promoting lipid deposition,potentially exerting its biological function through ceRNA network.
