[目的]探究拟杆菌(Bacteroides)早期干预对大肠杆菌攻毒雏鸡肠道健康的影响,为促进绿色健康养殖提供新思路.[方法]以150只初出壳健康爱拔益加(arbor acres,AA)雄性雏鸡为研究对象,随机分为对照组(第1日龄—第8日龄每天灌服生理盐水0.5 mL)、大肠杆菌组(第1日龄—第7日龄每天灌服生理盐水0.5 mL,第8日龄灌服禽致病性大肠杆菌菌悬液0.5 mL进行攻毒)和拟杆菌组(第1日龄—第7日龄每天灌服拟杆菌菌悬液0.5 mL,第8日龄灌服禽致病性大肠杆菌菌悬液0.5 mL进行攻毒).在攻毒后的第3天(第10日龄)和第14天(第21日龄),每组随机挑选8只鸡进行屠宰采样,对其生长性能、血清生化和抗氧化指标、血清炎症因子指标、肠道形态、肠道屏障相关基因的表达及肠道菌群进行测定.[结果]拟杆菌早期干预对大肠杆菌攻毒雏鸡生长性能和免疫器官指数的影响并不显著,但能显著提高雏鸡总抗氧化能力和血清中的超氧化物歧化酶酶活力,改善雏鸡血清中炎症因子水平;同时显著提高攻毒后第3天雏鸡空肠OCLN、CLDN3、ZO1、ZO2 mRNA相对表达量,以及回肠CLDN3、ZO1、ZO2 mRNA相对表达量,显著提高攻毒后第14天雏鸡空肠和回肠OCLN、CLDN3、ZO1、ZO2 mRNA相对表达量.拟杆菌早期干预使雏鸡盲肠菌群结构和相对丰度发生显著变化,雏鸡盲肠中拟杆菌属的相对丰度提升,而大肠杆菌志贺氏菌属(Escherichia_Shigella)的相对丰度下降.[结论]拟杆菌早期干预显著改善了大肠杆菌攻毒雏鸡免疫调节能力,并改变雏鸡肠道菌群的定植规律,有效缓解大肠杆菌感染导致的机体损伤.

[Objective]This study aimed to investigate the alleviating effects of early intervention with Bacteroides on intestinal health in chickens challenged with Escherichia coli(E.coli),providing new insights for sustainable and healthy poultry farming.[Method]A total of 150 newly hatched healthy arbor acres(AA)male chicks were randomly divided into three groups:(1)Control group(gavaged daily with 0.5 mL physiological saline from day 1 to day 8);(2)E.coli group(gavaged daily with 0.5 mL physiological saline from day 1 to day 7,followed by a challenge with 0.5 mL avian pathogenic E.coli suspension on day 8);and(3)Bacteroides group(gavaged daily with 0.5 mL Bacteroides sus-pension from day 1 to day 7,followed by a challenge with 0.5 mL avian pathogenic E.coli suspension on day 8).At 3 days(10 days of age)and 14 days(21 days of age)post-challenge,8 chickens per group were randomly selected for slaughter and sampling.Growth performance,serum biochemical and antioxidant indices,serum inflammatory cytokines,intestinal morphology,expression of intestinal barrier-related genes,and gut microbiota composition were analyzed.[Result]Early intervention with Bacteroides showed no significant effects on growth performance or immune organ indices in E.coli challenged chickens.However,it significantly enhanced total antioxidant capacity and serum supero-xide dismutase activity,improved serum inflammatory cytokine levels.The relative mRNA expression levels of OCLN,CLDN3,ZO1,and ZO2 in the jejunum,as well as CLDN3,ZO1,and ZO2 in the ileum,were significantly upregulated on day 3 post-challenge.On day 14 post-challenge,the relative mRNA expression levels of OCLN,CLDN3,ZO1,and ZO2 in both the jejunum and ileum were sig-nificantly increased.Additionally,Bacteroides intervention altered cecal microbiota composition,increasing the relative abundance of Bacteroides and reducing Escherichia_Shigella abundance.[Conclusion]Early intervention with Bacteroides significantly improved immune regulation in E.coli challenged chickens and reshaped gut microbiota colonization patterns,effectively alleviating pathogen-induced physiological damage.
