LHPP过表达对胰腺癌的抑制作用OA
The inhibitory effect of LHPP over-expression on pancreatic cancer
目的 探讨磷酸赖氨酸磷酸组氨酸无机焦磷酸磷酸酶(LHPP)过表达对胰腺癌的抑制作用.方法 回顾性收集2022年6月至2025年6月于内蒙古自治区人民医院就诊的18例胰腺癌患者的癌组织及配对癌旁组织样本,通过RT-qPCR检测其中LHPP mRNA表达.选取人胰腺癌细胞系Capan-2、Panc-1及正常胰腺细胞系HPDE6-C7,通过RT-qPCR和Western blotting法检测LHPP mRNA和蛋白的表达.构建并提取LHPP过表达质粒,采用脂质体瞬时转染法将其导入Capan-2和Panc-1细胞(过表达组),建立LHPP过表达细胞株,并以空载体转染细胞为对照(空载组).通过CCK-8法检测细胞增殖,Transwell实验评估细胞侵袭能力,划痕实验分析细胞迁移能力,流式细胞术检测细胞凋亡率.结果 在胰腺癌组织中,LHPP的mRNA表达水平较癌旁组织低(P<0.01).在胰腺癌细胞系Capan-2及Panc-1中,LHPP的mRNA和蛋白表达水平显著低于正常细胞HPDE6-C7(P<0.05).转染LHPP过表达质粒后,过表达组Capan-2及Panc-1中LHPP的mRNA和蛋白表达显著上调.CCK-8实验显示,转染LHPP后第3、4、5天Capan-2及Panc-1细胞过表达组相对细胞存活率均显著低于空载组(P<0.01).Transwell实验显示,转染LHPP后Capan-2及Panc-1细胞过表达组细胞数显著少于空载组(P<0.001).细胞划痕试验结果表明,转染LHPP后Capan-2及Panc-1细胞过表达组细胞迁移率显著低于空载组(P<0.05).流式细胞术结果表明,在瞬时转染LHPP后Capan-2及Panc-1细胞过表达组细胞凋亡率显著高于空载组(P<0.05).结论 LHPP过表达可显著抑制胰腺癌细胞增殖、迁移及侵袭,促进胰腺癌细胞凋亡.
Objective To investigate the inhibitory effect of phospholysine phosphohistidine inorganic pyrophosphate phosphatase(LHPP)over-expression on pancreatic cancer.Methods Cancer tissues and paired adjacent tissues from 18 pancreatic cancer patients admitted to Inner Mongolia People's Hospital between June 2022 and June 2025 were collected retrospectively,and LHPP mRNA expression was detected via RT-qPCR.Human pancreatic cancer cell lines Capan-2 and Panc-1,along with the normal pancreatic cell line HPDE6-C7,were selected to examine LHPP mRNA and protein expression using RT-qPCR and Western blotting.The LHPP over-expressed plasmid was constructed and extracted,then transfected into Capan-2 and Panc-1 cells using the liposome method to establish LHPP over-expressed cell lines(over-expressed group),with empty vector-transfected cells serving as controls(empty vector group).Cell proliferation was detected by CCK-8 assay,invasion ability was evaluated using Transwell assay,migration was analyzed via wound healing assay,and apoptosis was measured by flow cytometry.Results In pancreatic cancer tissues,LHPP mRNA expression was significantly lower than in paired adjacent tissues(P<0.01).The mRNA and protein expression levels of LHPP in pancreatic cancer cell lines Capan-2 and Panc-1 were significantly lower than in the normal pancreatic cell line HPDE6-C7(P<0.05).After transfection with the LHPP over-expressed plasmid,both mRNA and protein levels of LHPP were significantly upregulated in Capan-2 and Panc-1 cells.CCK-8 assay showed that on day 3,4,and 5 after transient transfection with LHPP,the relative cell viability in the over-expressed groups was significantly lower than that in the empty vector group(P<0.01).Transwell assay results indicated that the number of invading cells in the LHPP over-expressed group was significantly reduced compared with the empty vector group(P<0.001).Wound healing assay demonstrated that cell migration was significantly inhibited in the LHPP over-expressed group(P<0.05).Flow cytometry analysis revealed that the apoptosis rate was significantly higher in the LHPP over-expressed Capan-2 and Panc-1 cells than that in the empty vector control group(P<0.05).Conclusion Over-expression of LHPP can significantly inhibit the proliferation,migration,and invasion of pancreatic cancer cells and promote their apoptosis.
