[目的]探明香芹酚与百里香酚及其衍生物对杜仲灰斑病致病菌细极链格孢菌(Alternaria tenuis Nees)的生物活性,明确其对该致病菌的抑菌作用机制,为开发基于单萜酚类化合物及其衍生物的绿色高效杀菌剂防控杜仲灰斑病提供理论依据.[方法]以细极链格孢菌悬浮液为空白对照,以百菌清为阳性对照,分别设 5种不同浓度的香芹酚、百里香酚、乙酰化香芹酚和乙酰化百里香酚处理,测定杜仲灰斑病菌的菌丝生长速率、菌丝完整性和细胞膜通透性.[结果]香芹酚、百里香酚、乙酰化香芹酚和乙酰化百里香酚对杜仲灰斑病致病菌细极链格孢菌均有抑制活性,且抑制率均随浓度升高呈上升趋势,具有剂量依赖性,分别在浓度 130 mg/L、300 mg/L、130 mg/L、300 mg/L抑制率最高,分别为 90.09%、86.38%、85.14%、85.38%;培养 6 d的EC50 为 48.662~164.080 mg/L.香芹酚和百里香酚处理的杜仲灰斑病菌菌丝形态有扭曲变形和断裂情况,而各浓度乙酰化香芹酚和乙酰化百里香酚处理的菌丝形态未发现明显变化.香芹酚、百里香酚、乙酰化香芹酚及乙酰化百里香酚处理杜仲灰斑病病原菌,可增强菌丝的K+含量和细胞膜通透性,导致内溶物外泄,且随处理时间延长细胞膜通透性下降;百菌清处理对增强细胞膜通透性的效果不明显.[结论]香芹酚、百里香酚及其乙酰化衍生物对杜仲灰斑病致病菌细极链格孢菌均有抑菌活性,在绿色环保安全性上较百菌清表现更佳,具有较好的开发潜力.

[Objective]The biological activity of carvacrol,thymol and their derivatives against Alternaria tenuis causing gray spot disease of Eucommia ulmoides was investigated to clarify their antibacterial mechanisms against the pathogen,and provide a theoretical basis for development of green and efficient fungicides based on monoterpene phenols and their derivatives to effectively prevent and control gray spot disease of E.ulmoides.[Method]The effects of 4 compounds including carvacrol,thymol,acetylated carvacrol and acetylated thymol with 5 different concentrations on mycelial growth rate,mycelial integrity and cell membrane permeability of gray spot disease of E.ulmoides were determined by taking A.tenuis suspension and chlorothalonil as the blank control and positive control respectively.[Result]Carvacrol,thymol,acetylated carvacrol and acetylated thymol all had inhibitory activity against A.tenuis causing gray spot disease of E.ulmoides,and the inhibitory rate was on the rise with increase of fungicide concentration.The inhibitory rate of 130 mg/L carvacrol,300 mg/L thymol,130 mg/L acetylated carvacrol and 300 mg/L acetylated thymol against A.tenuis was 90.09%,86.38%,85.14%and 85.38%and their EC50 values after 6 d culture ranged from 48.662 to 164.080 mg/L.The mycelial morphology of A.tenuis treated with carvacrol and thymol showed distortion and fracture status,but the mycelial morphology of A.tenuis treated with acetylated carvacrol and acetylated thymol under 5 different concentrations was not changed obviously.Carvacrol,thymol,acetylated carvacrol and acetylated thymol treatment increased K+content and cell membrane permeability of mycelial,which insulted in release of intracellular substances in mycelial and the cell membrane permeability of mycelial decreased with extension of treatment time.Chlorothalonil had a little effect on increase of cell membrane permeability.[Conclusion]Carvacrol,thymol and their acetylated derivatives all have the inhibitory activity against A.tenuis,causing gray spot disease of E.ulmoides and are better than chlorothalonil in safety of green and environmental protection,which indicates that these compounds have a better development potential.
