[目的]分析金钗石斛叶片 2个发育阶段差异表达基因的表达特征,为研究金钗石斛为代表的兰科植物叶片生长发育提供参考.[方法]以金钗石斛为试验材料,采用Illumina高通量测序技术对金钗石斛叶片进行测序,分析金钗石斛叶片 2个发育阶段的转录组数据和基因表达特征,识别每个发育时期的基因表达模式.[结果]通过转录组测序共获得 126 981条单基因,平均长度为 1 035 bp.识别金钗石斛叶片 2个发育阶段的差异表达基因,有 313个上调表达和 757个下调表达.对差异表达基因进行功能聚类发现,代谢途径和次生代谢物的生物合成等途径可能与金钗石斛叶片发育密切相关.此外,鉴定出 9 917条编码转录因子的单基因,从属于 57个基因家族.其中,有 335个MYB转录本,在 2个发育阶段的叶片表达谱发现,2个MYB基因上调表达和 5个MYB基因下调表达.[结论]对金钗石斛叶片 2个发育阶段的转录组比较分析,下调基因数量显著多于上调基因,并鉴定出MYB、MADS、NAC和WRKY转录因子家族成员.

[Objective]The expression characteristics of differentially expressed genes in Dendrobium nobile leaves at two development stages were analyzed to provide a reference for growth and development of D.nobile.[Method]The transcriptome data and gene expression characteristics of D.nobile leaves at two development stages were analyzed by using Illumina high-throughput sequencing technology to identify the gene expression pattern of D.nobile leaves at different development stage.[Result]A total of 126 981 single-genes with an average length of 1 035 bp were obtained by transcriptome sequencing.The differentially expression genes with 313 up-regulated genes and 757 down-regulated genes were identified in D.nobile leaves at two different developmental stages.The functional clustering of differentially expressed genes showed that the metabolic pathways and biosynthesis of secondary metabolites may be closely related to D.nobile leaf development.In addition,9 917 single-genes of encoding transcription factors were identified and subject to 57 gene families.Among them,there were 335 MYB transcripts.Two up-regulated and five down-regulated MYB genes were identified in D.nobile leaves by analyzing their expression profiles at two developmental stages.[Conclusion]The transcriptome comparative analysis of D.nobile leaves at two developmental stages shows that the number of down-regulated genes is significantly higher than that of up-regulated genes,and members of MYB,MADS,NAC and WRKY transcription factor families are identified.
