[目的]探明水稻OsABF基因家族成员的理化特性和结构特征并验证其表达模式,为解析水稻OsABF基因家族的生物学功能奠定基础.[方法]采用生物信息学鉴定水稻OsABF家族成员,分析其系统进化、基因结构、保守基序与共线性、启动子元件和不同组织的表达模式,通过qRT-PCR技术分析OsABF基因在不同非生物胁迫下的表达模式.[结果]从水稻基因组中鉴定出 11个 OsABF基因,命名为OsABF1~OsABF11,蛋白序列长度为 244~388个氨基酸,平均长度为 332个氨基酸;OsABF蛋白大小为25.48~41.48 kDa,平均大小为 35.40 kDa;等电点为 5.30~9.56.11个OsABF蛋白被分为 2个亚家族,其中,A组包括OsABF4、OsABF7、OsABF8、OsABF10、OsABF1,B组包括OsABF1、OsABF2、OsABF3、OsABF5、OsABF6、OsABF9.OsABF在 2个亚家族中相对保守,片段复制是驱动水稻OsABF基因家族进化的主要因素,纯化选择在该基因家族进化中发挥重要作用.OsABF基因上游的启动子区鉴定出8种植物激素响应、6种植物生长发育调控、5种非生物/生物胁迫响应的顺式作用元件,所有OsABF启动子序列均含ABRE顺式作用元件.表达谱分析显示,A组中OsABF基因在各组织中均有表达,且被干旱、低温、ABA和JA等非生物胁迫诱导表达.[结论]水稻 11个OsABF蛋白被分为A、B组亚家族,且功能相对保守,A组成员OsABF4、OsABF10在水稻生长发育和非生物胁迫响应中发挥重要作用.

[Objective]The physicochemical properties,structure characteristics and expression pattern of OsABF gene family members in rice were explored and verified to lay a foundation for biological functions of rice OsABF gene family.[Method]The members of rice OsABF family were identified by bioinformatics,and their phylogenetic relationships,gene structure,conserved motifs and collinearity,promoter elements and expression patterns in different tissues were analyzed.The expression patterns of OsABF genes under different abiotic stresses were analyzed by qRT-PCR technology.[Result]11 OsABF genes were identified from rice genome and named as OsABF1-OsABF11.The protein sequence length,size and isoelectric point of OsABF1-OsABF11 were 244-388 amino acids with an average length of 332 amino acids,25.48-41.48 kDa with an average of 35.40 kDa and 5.30-9.56,respectively.11 OsABF protein were divided into two subfamilies of A(OsABF4,OsABF7,OsABF8,OsABF10 and OsABF11)and B(OsABF1,OsABF2,OsABF3,OsABF5,OsABF6 and OsABF9).OsABF was relatively conserved in two subfamilies.The segmental duplication was the main factor driving evolution of rice OsABF gene family and the purifying selection played an important role in the evolution of rice OsABF gene family.The response of 8 plant hormones,regulation of 6 plant growing development and 5 cis-acting elements under abiotic and biological stresses were respectively identified from upstream promoters of OsABF genes,and the sequence of all OsABF promoters had ABRE cis-acting elements.The expression profile analysis showed that expression of most OsABF genes in subfamily A was in different tissues and induced by abiotic stresses such as drought,low temperature,ABA,and JA.[Conclusion]11 OsABF protein are divided into A and B subfamily with a relative conserved function.OsABF4 and OsABF10 in A subfamily play an important role in rice growing development and the response to abiotic stress.
