半夏提取物通过NLRP3/caspase-1通路对慢性阻塞性肺疾病大鼠肺损伤和气道炎症的影响OA
Effects of Pinellia ternata extract on lung injury and airway inflammation in rats with chronic obstructive pulmonary disease via NLRP3/caspase-1 pathway
目的 探讨半夏提取物通过核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)/胱天蛋白酶1(caspase-1)通路对慢性阻塞性肺疾病(COPD)大鼠肺损伤和气道炎症的影响.方法 将大鼠随机分为正常组、COPD组、半夏提取物低剂量组、半夏提取物高剂量组、半夏提取物高剂量+nigericin组,每组10只.检测各组大鼠肺功能、肺组织病理、炎症细胞浸润、肺泡灌洗液(BALF)和血清中炎性细胞因子水平.采用Western blotting检测肺组织中NLRP3/caspase-1通路蛋白的表达.结果 COPD组与正常组比较,半夏提取物高剂量+nigericin组与半夏提取物高剂量组比较,大鼠肺活量、呼气峰流值(PEF)均降低,吸气阻力(Ri),肺病理损伤评分,淋巴细胞、中性粒细胞和嗜酸性粒细胞数,肿瘤坏死因子α(TNF-α)、白细胞介素-1β(IL-1β)和白细胞介素-18(IL-18)水平,NLRP3和caspase-1蛋白相对表达水平均升高(P<0.05).半夏提取物低剂量组和高剂量组与COPD组比较,大鼠肺活量、PEF均升高,Ri,肺病理损伤评分,淋巴细胞、中性粒细胞和嗜酸性粒细胞数,TNF-α、IL-1β和IL-18水平,NLRP3和caspase-1蛋白相对表达水平均降低(P<0.05).半夏提取物高剂量组大鼠各病理指标的改善更明显.结论 半夏提取物可通过抑制NLRP3/caspase-1通路激活,减轻COPD大鼠肺损伤和气道炎症.
Objective To investigate the effect of Pinellia ternata extract on lung injury and airway inflammation in rats with chronic obstructive pulmonary disease(COPD)based on the nucleotide-binding oligomerization domain-like receptor protein 3(NLRP3)/caspase-1 pathway.Methods Rats were randomly assigned into normal,COPD,low-dose Pinellia ternata extract,high-dose Pinellia ter-nata extract,and high-dose Pinellia ternata extract+nigericin groups(n=10 in each group).Lung function,inflammatory cell infiltration,and the levels of inflammatory cytokines in the bronchoalveolar lavage fluid(BALF)and serum were evaluated,and a pathological exam-ination of lung tissue was conducted.Western blotting was used to detect the expression of NLRP3/caspase-1 pathway-related proteins in lung tissue.Results In the COPD group(compared with the normal group)and the high-dose of Pinellia ternata extract+nigericin group(compared with the high-dose Pinellia ternata extract group),the lung capacity and peak expiratory flow(PEF)of rats were decreased,while the inspiratory resistance(Ri),lung pathological injury score,lymphocyte,neutrophil and eosinophil counts,levels of tumor necrosis factor-α(TNF-α),interleukin(IL)-1β,and IL-18,and relative expression of NLRP3 and caspase-1 proteins were increased(P<0.05).Compared with the COPD group,the lung capacity and PEF of rats in in low-and high-dose Pinellia ternata extract groups were increased,while Ri,the lung pathological injury score,lymphocyte,neutrophil,and eosinophil counts,levels of TNF-α,IL-1β,and IL-18,and relative expression of NLRP3 and caspase-1 proteins were decreased(P<0.05).The high-dose Pinellia ternata extract showed stronger improve-ment effects on various pathological indicators of rats with COPD.Conclusion Pinellia ternata extract can alleviate lung injury and airway inflammation in rats with COPD by inhibiting the activation of the NLRP3/caspase-1 pathway.
