鸭瘟病毒野毒株的分离鉴定和疫苗免疫试验OA
Isolation,Identification and Vaccine Immunity Testing of a Wild Type Duck Plague Virus Strain
为了解2024年1月江苏省某种鸭场樱桃谷种鸭发病是否与鸭瘟病毒变异有关,本试验对病死鸭的肝脏、脾脏、肠道、食管和气管组织进行病毒分离培养,并基于长独特区域2(UL2)基因和长区开放阅读框5(LORF5)基因对分离毒株进行PCR扩增,同时进行动物回归试验、毒力测定和疫苗免疫效果评价.结果显示,成功分离到1株鸭瘟病毒,命名为QL240109株;该分离毒株的UL2基因片段长度与弱毒株相同,LORF5基因片段长度与强毒株相同;动物回归试验结果显示,本试验分离毒株对健康易感樱桃谷肉鸭具有较强的致死性;毒力测定结果显示,本试验分离毒株毒力低于我国经典强毒株CVCC AV1221株;疫苗免疫效果评价结果显示,鸭瘟活疫苗对该分离毒株的免疫保护率为75%~100%.结果表明,与我国弱毒疫苗株和经典强毒株相比,本试验分离毒株的基因组结构发生了变异,致死性强但毒力低于经典强毒株,目前鸭瘟活疫苗可以提供有效保护.本试验结果为养殖场防控该鸭瘟病毒变异株提供了参考依据.
To determine whether the outbreak in Cherry Valley breeder ducks in a farm in Jiangsu Province in January 2024 was related to a variant of duck plague virus,tissues from the liver,spleen,intestine,esophagus,and trachea of dead ducks were collected for virus isolation and culture.The isolated strain was subjected to PCR amplification based on the unique long region 2(UL2)gene and long open reading frame 5(LORF5)gene,as well as animal regression experiments,virulence determination,and vaccine immunization evaluation.The results showed that one duck plague virus strain was successfully isolated and designated QL240109.The UL2 gene fragment length of this strain was consistent with that of the attenuated strain,whereas the LORF5 gene fragment length was consistent with that of the virulent strain.Animal regression experiments demonstrated that the isolated strain was highly lethal to susceptible Cherry Valley ducks.Virulence determination indicated that the strain had lower virulence than the classical Chinese virulent strain CVCC AV1221.Vaccine efficacy evaluation revealed that the duck plague live vaccine provided 75%-100% protection against the isolated strain.These findings suggest that,compared with the attenuated vaccine strain and classical virulent strains in China,the isolated strain underwent genomic variation,exhibited strong lethality but lower virulence than classical virulent strains,and could still be effectively controlled by duck plague live vaccines.The results provide a reference for the prevention and control of this mutant strain of duck plague virus in duck farms.
