2024-2025年河北省犊牛源腹泻大肠杆菌流行病学调查OA
Epidemiological Investigation of Diarrheagenic Escherichia coli from Calves in Hebei Province from 2024 to 2025
为了解河北省犊牛腹泻大肠杆菌的流行病学特征,本试验于2024-2025年采集240份犊牛腹泻粪便样品进行大肠杆菌的分离鉴定,并对其毒力基因、药物敏感性、耐药基因和生物被膜形成能力进行检测,同时进行系统进化群分型和多位点序列分型(MLST).结果显示,240份犊牛腹泻粪便样品中分离鉴定出214株大肠杆菌,分离率为89.17%;毒力基因检测结果显示,分离菌株的毒力基因crl(100%)、fliC(92.99%)和ompA(87.85%)检出率较高,而tsh、afa和traB基因未检出;系统进化群分型结果显示,分离菌株以B1群(54.21%)和A群(33.18%)为主;药敏试验结果显示,分离菌株对链霉素(65.89%)和环丙沙星(50.47%)耐药率较高,多重耐药率达51.40%,且24.77%的菌株对10种以上抗生素耐药;耐药基因检测发现,四环素类耐药基因tet(A)(31.78%)、β内酰胺类耐药基因blaTEM(28.97%)和磺胺类耐药基因sul2(28.97%)的检出率较高;生物被膜形成能力检测结果显示,60.75%的分离菌株可形成生物被膜,其中B2群菌株均具有生物被膜形成能力.MLST结果显示,选取的40株多重耐药菌株中有38种序列型(ST),包括15种新型ST,优势ST为ST20和ST1140.本试验为河北省奶牛和肉牛养殖场针对性制定由大肠杆菌引起犊牛腹泻病的防控策略提供了理论依据.
To investigate the epidemiological characteristics of Escherichia coli(E.coli)associated with calf diarrhea in Hebei Province,a total of 240 fecal samples from diarrheic calves were collected between 2024 and 2025 for E.coli isolation and identification.The isolates were analyzed for virulence genes,antimicrobial susceptibility,antimicrobial resistance genes,and biofilm-forming ability,along with phylogenetic group classification and multilocus sequence typing(MLST).The results showed that 214 E.coli isolates were recovered,with an isolation rate of 89.17%.The predominant virulence genes were crl(100%),fliC(92.99%),and ompA(87.85%),while tsh,afa,and traB were not detected.Phylogenetic analysis revealed that most isolates belonged to groups B1(54.21%)and A(33.18%).Antimicrobial susceptibility testing indicated high resistance rates to streptomycin(65.89%)and ciprofloxacin(50.47%),with 51.40%of isolates showing multidrug resistance,and 24.77%resistant to more than 10 antibiotics.The most frequently detected antimicrobial resistance genes were tetracycline resistance gene tet(A)(31.78%),β-lactam resistance gene blaTEM(28.97%),and sulfonamide resistance gene sul2(28.97%).Biofilm-forming ability assays showed that 60.75%of isolates were capable of forming biofilms,and all B2 group strains exhibited biofilm-forming ability.MLST analysis of 40 multidrug resistance isolates identified 38 distinct sequence type(ST),including 15 novel ST,with ST20 and ST1140 as the predominant types.This study provides a theoretical basis for developing targeted prevention and control strategies for calf diarrhea caused by E.coli in dairy and beef cattle farms in Hebei Province.
