目的:探讨miRNA-133调节人源线粒体内膜转位酶8a(hTim8a)对高糖诱导的肾小管上皮细胞损伤的影响.方法:体外培养大鼠肾小管上皮细胞HK-2,将HK-2细胞分为CK组、高糖组、高糖+inhibitor-NC组、高糖+miRNA-133 inhibitor组;qRT-PCR检测细胞miRNA-133和TIMMA基因表达;MTT法检测细胞增殖;流式细胞术检测细胞凋亡率;生化法检测ROS和ATP水平;ELISA试剂盒检测LDH、TNF-α、IL-6和IL-10水平;商品化试剂盒检测MDA、SOD、CAT水平;Western blot检测细胞中cleaved caspase-3、hTim8a、mt-COX、PGC-1、PRC蛋白表达.结果:与CK组相比,高糖组HK-2细胞miRNA-133表达、细胞凋亡率、ROS、LDH、TNF-α、IL-6水平、MDA含量、cleaved caspase-3蛋白表达显著升高,TIMMA mRNA表达、A490(24 h、48 h)值、ATP、IL-10水平、SOD和CAT活性、hTim8a、mt-COX、PGC-1、PRC蛋白水平显著降低(P<0.05);与高糖+inhibitor-NC组相比,高糖+miRNA-133 inhibitor组HK-2细胞miRNA-133表达、细胞凋亡率、ROS、LDH、TNF-α、IL-6水平、MDA含量、cleaved caspase-3蛋白表达显著降低,TIMMA mRNA表达、A490(24 h、48 h)值、ATP、IL-10水平、SOD和CAT活性、hTim8a、mt-COX、PGC-1、PRC蛋白水平显著升高(P<0.05).结论:沉默miRNA-133表达可上调hTim8a表达,减轻高糖诱导的HK-2细胞炎症、氧化应激和细胞凋亡,进而提高细胞活性和线粒体功能,减轻细胞损伤.

Objective:To investigate the effect of miRNA-133 on high glucose induced renal tubular epithelial cell injury by regulating human translocase of inner mitochondrial membrane 8a(hTim8a).Methods:Rat renal tubular epithelial cells HK-2 were cultured in vitro and divided into CK group,high glucose group,high glucose+inhibitor-NC group,and high glucose+miRNA-133 in-hibitor group.qRT-PCR was used to detect expressions of miRNA-133 and TIMMA genes in cells.MTT method was used to detect cell proliferation.Flow cytometry was used to detect cell apoptosis rate.Biochemical methods were used to detect ROS and ATP levels.ELI-SA kits were used to detect LDH,TNF-α,IL-6 and IL-10 levels.Commercialized reagent kits were used to detect MDA,SOD and CAT levels.Western blot was used to detect expressions of cleaved caspase-3,hTim8a,mt-COX,PGC-1 and PRC proteins in cells.Results:Compared with CK group,miRNA-133 expression,apoptosis rate,levels of ROS,LDH,TNF-α,IL-6,MDA content,and expression of cleaved caspase-3 protein in HK-2 cells in high glucose group were greatly increased,while the expression of TIMMA mRNA,A490(24 h,48 h)values,levels of ATP and IL-10,activities of SOD and CAT,protein levels of hTim8a,mt-COX,PGC-1,PRC were greatly reduced(P<0.05).Compared with high glucose+inhibitor-NC group,miRNA-133 expression,apoptosis rate,levels of ROS,LDH,TNF-α,IL-6,MDA content,and protein expression of cleaved caspase-3 in HK-2 cells in high glucose+miRNA-133 inhibitor group were greatly reduced,TIMMA mRNA expression,A490(24 h,48 h)values,levels of ATP and IL-10 levels,activities of SOD and CAT,protein levels of hTim8a,mt-COX,PGC-1,PRC were greatly increased(P<0.05).Conclusion:Silencing expression of miRNA-133 can up-regulate the expression of hTim8a,alleviate high-glucose induced inflammation,oxidative stress,and apoptosis in HK-2 cells,thereby improving cell activity and mitochondrial function,and reducing cell injury.
