耐受性树突状细胞对胶原诱导性关节炎大鼠肠道菌群结构的影响OA
Effect of tolerogenic dendritic cells on gut microbiota structure in collagen-induced arthritis rats
目的:探究耐受性树突状细胞(tolDC)对胶原诱导性关节炎(CIA)大鼠肠道菌群结构的影响.方法:实验动物随机分为Control组、CIA组、tolDC组.CIA组与tolDC组大鼠均注射牛Ⅱ型胶原乳剂建立CIA模型;tolDC组大鼠于初次免疫第21天尾静脉输注tolDC进行治疗;每周记录各组大鼠关节炎评分及足跖厚度;第35天处死大鼠,留取标本进行检测.HE染色检测各组大鼠踝关节病理学变化,16S rDNA检测肠道菌群α-多样性、β-多样性、物种组成与物种差异.结果:与Control组大鼠相比,CIA组大鼠关节炎评分及足跖厚度增加,滑膜组织异常增生,滑膜炎症细胞浸润,并伴有血管翳产生,骨及软骨损伤,肠道菌群多样性指数Chao1、Shannon降低,但差异无统计学意义,厚壁菌门丰度降低,Clostridioides和Muribaculum丰度增加;tolDC治疗后CIA大鼠关节炎评分及足跖厚度降低,滑膜炎症细胞浸润、滑膜组织异常增生、血管翳产生、骨及软骨损伤等病理变化得到缓解,Chao1指数降低(P<0.01),厚壁菌门、乳杆菌(Lactobacillus)、Lachnoclostridium、GCA-900066575、Dubosiella、Pepto-coccus、Acetatifactor、Butyricicoccus等有益菌丰度上调.结论:tolDC可缓解CIA大鼠关节炎症,改善肠道菌群结构.
Objective:To investigate effect of tolerogenic dendritic cells(tolDC)on gut microbiota structure of rats with colla-gen-induced arthritis(CIA).Methods:Experimental animals were randomly divided into Control group,CIA group and tolDC group.CIA group and tolDC group were immunized with bovine type Ⅱ collagen solution to establish CIA model.TolDC group was treated with tolDC via tail vein on 21th day after initial immunization.Arthritis score and thickness of paws in each group of rats were mea-sured weekly.On the 35th day,rats were sacrificed and specimens were taken for testing.HE staining was performed on ankle joint to evaluate pathological changes in each group of rats.16S rDNA sequencing was used to detect changes in gut microbiota structure of rats in each group,including alpha diversity,beta diversity,species composition and species difference analysis.Results:Compared with Control group,arthritis score and thickness of paws in CIA group were increased,exhibited abnormal proliferation and extensive infiltration of inflammatory cells,as well as formation of pannus and destruction of bone and cartilage,intestinal flora index of Chao1 and Shannon were decreased,but difference was not significant,abundance of Firmicutes was decreased,abundance of Clostridioides and Muribaculum were increased.After tolDC treatment,arthritis score and thickness of paws of CIA rats were decreased,infiltration of inflammatory cells,abnormal hyperplasia of synovial tissue,formation of pannus and injury of bone and cartilage were alleviated,intestinal flora index of Chao1 was decreased(P<0.01),abundance of Firmicutes,Lactobacillus,Lachnoclostridium,GCA-900066575,Dubosiella,Peptococcus,Acetatifactor and Butyricicoccus were increased.Conclusion:TolDC alleviates joint injury in CIA rats and influences gut microbiota structure.
